本发明涉及基因工程技术领域,具体地说涉及一类表达于日本血吸虫表皮的蛋白质、其编码核苷酸和编码氨基酸序列以及其作为药物靶点和疫苗的作用。
背景技术:
目前研究表明,血吸虫病由血吸虫的感染引起,在世界上有76个国家,受感染者约1.5亿。致病的血吸虫有3种,既日本血吸虫(Schistosoma japonicum),曼氏血吸虫(S.mansoni)和埃及血吸虫(S.haematobium)。在我国流行的是日本血吸虫。
研制新的治疗血吸虫病和抗血吸虫的药物,首先必须找到合适的药物作用靶点。血吸虫的表皮是宿主和血吸虫相互作用的界面。而组成血吸虫表皮的蛋白质暴露在这个界面上,对小分子的化学药物和大分子的抗体药物而言都是最理想的药物作用靶点。已经有研究表明治疗血吸虫的化学药物——吡喹酮的作用靶点就是存在于血吸虫的表皮。另外一些已知的血吸虫表皮蛋白,例如GST28,paramyosin,已经用于疫苗研究和应用。
发明内容
本发明要解决的技术问题是提供一类表达于日本血吸虫表皮的蛋白质、其编码核苷酸、及编码氨基酸序列日本血吸虫表皮蛋白,作为可能的药物作用靶点和候选疫苗方面的作用。
为解决上述技术问题,本发明提供一种DNA序列,它具有序列表中SEQ IDNO.1~SEQ ID NO.20所述序列中任一序列的碱基序列;或者它是在中度严紧条件下与序列表中SEQ ID NO.1~SEQ ID NO.20所示的序列中任一条序列杂交的序列;或者它是与序列表中SEQ ID NO.1~SEQ ID NO.20所示的序列中任一条序列有至少70%同源性的序列。
为解决上述技术问题,本发明还提供一种氨基酸序列,它具有序列表中SEQID NO.21~SEQ ID NO.40所述序列中任一序列的氨基酸序列;或者它是与序列表中SEQ ID NO.21~SEQ ID NO.40所示的序列中任一条序列有至少70%同源性的序列;或者它是包含有序列表中SEQ ID NO.21~SEQ ID NO.40所示的序列中任一条序列的氨基酸序列。
本发明由于通过大规模的蛋白质组学研究,发现了20个日本血吸虫表皮蛋白质,这些蛋白质,都是可能的药物作用靶点和候选疫苗。血吸虫病的流行给人民身体健康和经济发展都造成巨大的损失,因此,防治血吸虫病、开发研制治疗血吸虫病和抗血吸虫的药物具有巨大的社会和经济效益。

具体实施例方式
下面结合附图和具体实施例对本发明作进一步详细的说明。
实施例1日本血吸虫表皮蛋白的分离和鉴定实验1.日本血吸虫表皮的分离纯化血吸虫表皮的纯化按照文献报道的Triton-x-100提取方法进行(参见Oaks,J.A.等人,寄生虫学杂志,69(3)519-533(1983)。)。表皮蛋白从血吸虫雄虫、雌虫、雌雄混合成虫和童虫分离。虫体用Krebs’-Ringer’s,Tris-马来酸盐-缓冲盐液(KRTM)洗3次,再用KRTM/0.2%Triton X-100溶液悬浮。这个液体和虫体的混和物置于冰水浴中,在摇床上摇动10分钟。然后,装有液体和虫体的混和物的试管在涡旋混合器上混旋几次,然后再放回冰水浴中,静置使虫体下沉。用吸管吸出上部液体,放置到另一个新管中。这个试管在离心机上用2500×g的离心力,离心15分钟。得到的沉淀就是血吸虫的表皮。Krebs’-Ringer’s溶液的配方是0.15M NaCl,6.2mM KCl,1.2mM Na2HPO4,1.2mM MgSO4,10mM Glucose and 20mM Tris-Hepes,pH7.4。Tris maieate缓冲溶液的配方是0.2M Tris and 0.2MMaleicanhyclride,pH 7.4。
2.日本血吸虫表皮蛋白的电泳分离日本血吸虫表皮用裂解剂裂解,蛋白溶液用SDS-聚丙烯酰胺凝胶电泳分离,条带切割后再做第二维的在线毛细液相色谱分离以及质谱鉴定。软件分析得到鉴定蛋白。分离好的血吸虫表皮,用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)缓冲液悬浮,用超声波促进组织细胞裂解和蛋白溶解,超声波裂解方法为超声1秒,暂停3秒,总时间2分钟。并用100℃加热5分钟以使蛋白质变性。然后这些表皮蛋白混合物用SDS-PAGE分离,分离方法按照普通的SDS凝胶电泳方法进行。胶上电泳泳道用刀切割下来,再用刀均等分割成8-12份。接下来做蛋白的胶内酶解。
3.日本血吸虫表皮蛋白的胶内酶解过程胶内酶解参照文献(Shevchenko,A.等人,化学年报,68850-858(1996)。)的方法进行。蛋白胶块用刀切碎,每个小块大小约为1立方毫米。加入100mMNH4HCO3,洗2次;转入到试管中;加入10mM二硫苏糖醇(dithiothreitol,DTT)/100mM NH4HCO3,56度放置反应1小时;冷却到室温,离心吸去DTT;加入等体积的55mM碘乙酰胺iodoacetamide/100mM NH4HCO3,室温/黑暗反应45分钟;吸去iodoacetamide,加入100mM NH4HCO3,洗2次;去掉NH4HCO3,加入乙腈缩胶;去掉乙腈,真空抽干;加入100mM NH4HCO3,胶膨胀,去掉NH4HCO3,加入乙腈缩胶;去掉乙腈,抽干;加入含Trypsin的消化缓冲液(50mM NH4HCO3/12.5ng/ul trypsin)冰上放置45min;去掉多余酶液,加入50mM NH4HCO3,以刚覆盖胶为准;37度,过夜酶切;酶切结束后,从试管中吸出上清液到新管子中,胶中加入60%ACN/0.5%HAC,在水浴中超声处理15min,再重复一次;吸出上清液到新管子中,在原管子中再加入60%ACN/0.5%HAC,37度40min,吸出上清液,与前面的上清液合并,真空抽气去ACN。
4.日本血吸虫表皮蛋白酶解肽段的在线色谱分离和质谱鉴定酶解产生的肽混合物,用真空将溶液体积抽减到30μl。之后,每个组分的肽混合物上样到一个反相捕获柱(C18,5μm,300,300μm i.d×5mm,LC Packings),在20μl/min的流速下脱盐。然后这个捕获柱和一个分析用的75μm×150mm C18柱(LC Packings)相连接,肽混合物就在200nl/min的流速下,从这个柱子上洗脱进入装有Protana NanoES的电喷雾离子源的QSTAR pulser i中。Agilent 1100毛细液相系统(Agilent Technologies)用来提供流动相A(0.5%乙酸/水)和流动相B(0.5%乙酸/乙腈),线性梯度是60分钟内5%B到50%B,30分钟内50%B到90%B,然后是在90%B停留15分钟。在分析柱后通过一个不锈钢的两通(Valco Instrument)连接一个10-μm直径的PicoTip纳喷雾头(New Objective),加上2500-3000伏的喷雾电压,以获得稳定的喷雾。MS/MS串联质谱谱图通过信息依赖的采集(IDA)和任务-循环增强来记录。在每个测定扫描中,选取3-6个信号最强、带2到3个电荷的离子,用滚动碰撞能量打碎,以获得串联质谱的信息。
5.日本血吸虫表皮蛋白的软件搜索鉴定用牛胰酶的自裂解片段如([M+H]+)1153.5741,2163.0569和2274.1600作为内标,或者以预先数据库搜索得到的得分高的肽段的分子量,来校正所有的质谱谱图。质谱搜索用的数据库,包括从8420个从EST cluster预测得到的血吸虫蛋白质序列,以及可能的污染蛋白质序列如牛胰酶的序列、宿主蛋白污染如人蛋白质序列、小鼠蛋白质序列、大鼠蛋白质序列、兔子蛋白质序列和角蛋白序列等。搜索软件是MASCOT(http://www.matrixscience.com/,MatrixScience)。搜索参数为最大漏切点,3个;monoisotopic;肽电荷数,+1/+2/+3;修饰,半胱氨酸carbamoylmethylation和methionine氧化。搜索结果合并后,用下列标准进行过滤筛选1.对每一个串联质谱谱图的搜索结果列表,只提取排位在第一个的结果,其他的则丢弃;2.搜索得到的肽段中如果有KR,KK,RK,RR等序列,则这个肽段丢弃;3.搜索得到的肽段,长度在8个氨基酸以上的保留,小于8个氨基酸的丢弃;4.重复打到的肽段,只计算其中分值最高的,把这些肽段分值加起来,做为蛋白的得分;5.蛋白的得分超过60,就认为是可信的结果。低分的蛋白,手工检查后保留或者丢弃。匹配到宿主或者牛胰酶、角蛋白的肽段,则丢弃。
实施例2日本血吸虫表皮蛋白的生物信息学分析和功能注释
1.同源性和相似性分析日本血吸虫表皮蛋白序列对GenBank(http://www.ncbi.nlm.nih.gov/)的非冗余数据库(Non-redundant,NR)做BLASTP比对;日本血吸虫表皮蛋白序列对Swissprot数据库做BLASTP同源性比对。根据比对结果得到的与已知其他物种或已知的血吸虫蛋白的序列相似性,做出日本血吸虫表皮蛋白的功能注释。
2.文献查检根据已经发表的文献报道,发现鉴定得到的20个表皮蛋白在虫体上的位置,有一些的确在血吸虫表皮上。而其他则是新发现的表皮蛋白。
举例说明,比如以下表皮分离和质谱鉴定得到的蛋白,根据文献报道,就是已知的表皮蛋白,而且有些蛋白已经有进行了疫苗和抗原研究。

21.7 kDa antigenFK506-binding protein 50(immunophilin)实施例3日本血吸虫表皮蛋白的实验验证为了进一步用实验的方法验证用蛋白质组学手段鉴定得到的血吸虫表皮蛋白,选取了一个Immunophilin(SjFKBP50,GenBank登录号为AY815389)做免疫荧光抗体原位杂交做蛋白定位分析。
实验方法血吸虫成虫用冷冻包埋剂(OCT)完全包埋,切成5μm厚的切片。用抗Immunophilin的抗体覆盖切片,在37℃反应60分钟。该抗体是用重组表达的Immunophilin免疫小鼠制备的单克隆抗体。随后切片用PBS洗涤,再加入FITC-偶联的兔抗鼠免疫球蛋白的抗体(1∶40稀释,用含有0.5mg/ml的Evans蓝的PBS稀释),置湿盒中37℃反应60分钟。染色结果用Leica DM-RB型荧光显微镜观察并拍照。
图一所示为免疫荧光抗体原位杂交实验结果,表皮T;肠道G;下表皮ST。绿色荧光部分是Immunophilin的信号。
该结果证明了Immunophilin确实分布在血吸虫成虫的表皮上。这说明前面表皮分离和质谱鉴定的结果是可信的。

本发明公开了一类在日本血吸虫表皮上表达的表皮蛋白,尤其公开了所述表皮蛋白的编码核苷酸和编码氨基酸。通过对其进行大规模的蛋白质组学研究,认为它们可能具有药物作用靶点和候选疫苗的作用。血吸虫病的流行给人民身体健康和经济发展都造成巨大的损失,因此,防治血吸虫病、开发研制治疗血吸虫病和抗血吸虫的药物具有巨大的社会和经济效益。
