肽聚糖 存在于原核生物细胞壁的大分子聚合物。其骨架是由N-乙酰葡萄糖胺(N-acetylglucosamine简写G)和N-乙酰胞壁酸(N-acetylmuramic acid简写M)通过β-1，4糖苷键交替相联而组成的线状聚糖链。M是在N－乙酰葡萄糖胺的C3位置上联结一个乳酰醚。就在M的乳酰基上,联结着一条由四个氨基酸残基组成的短肽链。短肽链的氨基酸组成因菌种而异，包括不常见的D－谷氨酸、D－丙氨酸、L-二氨基庚二酸和其他二氨基酸；其中L型和D型氨基酸交替排列。 由G和M组成的二糖以及联在M上的四肽是肽聚糖的基本结构单位（图1）。聚糖链的长度亦因菌种而异,短到9个二糖单位，长到170多个二糖单位；1个二糖单位长10.3埃，所以总长度可从 100埃到1700埃左右。各个四肽链之间有交联。在革兰氏阴性菌,多半是两条肽聚糖的肽链直接交联,交联的肽链占肽链总数的50％。而革兰氏阳性菌则是通过另一条肽桥交联，如：金黄色葡萄球菌的肽桥为五个甘氨酸组成的五肽，交联的肽链占肽链总数的 75～100％（图2）。立体的交联使肽聚糖形成多层次网状结构。 革兰氏阴性菌，因肽聚糖层次较少（如：大肠杆菌为3层，厚度约2～3纳米），交联程度较低,所以网状结构疏松；而革兰氏阳性菌，因肽聚糖层次较多（如：金黄色葡萄球菌约10层,枯草杆菌约20层,厚度可达10～50纳米），交联程度较高，网状结构紧密。这两类细菌在肽聚糖含量、厚度、层次和交联程度上的差别，再加上细胞壁其他组份上的不同，反映在染色性质，对溶菌酶敏感程度和物质透过性质等方面都有很大的不同。少数细菌，如：嗜盐菌，细胞壁结构特殊，不含肽聚糖。 青霉素、万古霉素、杆菌肽、磷霉素、环丝氨酸和持久霉素等几种抗菌素，分别作用于肽聚糖生物合成的不同步骤,抑制肽聚糖生物合成,从而抑制细菌生长。溶菌酶使肽聚糖的聚糖链断裂，破坏细胞壁结构，将细菌杀死。
