~(15)N分析样品制备~(15)N分析样品制备sample preparation for ~(15)N analysis  ‘SN分析样品制备(sample preparation for1,N analysis)使’SN示踪试验材料变成适于‘,N光谱分析或质谱分析的化学预处理和氮的转化的技术.无论是质谱分析或光谱分析都要求将样品中各种形态的氮转化为分子态氮。N:样品的制备分两步进行,第一步是将样品预处理或化学处理。大多数情况下经凯氏消化将各种形态氮转化为钱态氮;第二步是氮的转化,将按态氮或其他形态的氮转化为分子氮. 样品的化学预处理收集的植物材料和土壤样品必须烘干或风干、称重、磨碎,充分混合均匀,然后用不同的方法进行化学预处理。大多数情况下,采用凯氏法(Kjeldahl Method)将样品氮转化为钱态氮,然后与强碱作用,释放的氨由燕汽逐出,被标准盐酸或硫酸或2%翻酸吸收,滴定法或比色法测定氨的量。将蒸馏液酸化后燕发浓缩至少量体积,作为转化NZ用。 消化样品加浓硫酸和催化剂,经高温(358C)消煮.将各种形态氮转化为按态氮。经典的凯氏法以KMnO‘为催化剂,改良凯氏法改用硒、氧化汞、硫酸铜和过氧化氢等,而且加入硫酸盐以提高反应液温度。试验中推荐用·KZSO。:CuSO;:Se=100:10:z的混合催化剂于一般的植物和土壤样品。 蒸馏取一定量消化液于反应瓶中,加入强碱(10摩尔/升,Na()H),释放的氨由蒸汽逐出.以2%硼酸溶液吸收,加入甲基红一澳甲酚绿混合指示剂,用标准硫酸滴定。也可用标准硫酸或标准盐酸为吸收液,用标准碱液回滴剩余的酸。亚硝态氮样品用常规方法不能完全转化为钱,需要在消化之前对样品进行预处理。常用高锰酸钾一还原铁法。 天的转化将按态氮或其他各种形态的氮转化为分子氮。 列登伯格法(Rettenberg Method)将上述吸收液用1摩尔/升的HZso‘液酸化并浓缩的至小量体积,于真空中与碱性次澳酸钠反应,产生分子态氮: ZNH3+3Na0Br eeNZ+3H:0+3NaBr碱性次澳酸钠溶液注入Y形反应瓶的一侧份,含有按列登伯榕管通的样品溶液注入另一侧臂,委将Y形瓶连接到真空系统巫上。液氮冷冻除气后旋转Y统二一;~丫二,_.’_二’:”了,几’丁一瓶,使两液混合,反应产生 N2.再将NZ导入质谱仪的 离子室或封入光谱测t用的 放电管内,供’SN分析用.因 为此法中所用的样品是凯氏 消化一蒸馏以后的含NH.十 溶液,所以也称凯氏一列登 伯格法。
