HPLC六通阀进样器的使用及保养说明 六通阀进样器是高效液相色谱系统中最理想的进样器,它是由圆形密封垫(转子)和固定底座(定子)组成。美国Rheodyne公司的六通阀进样器最为通用,各大HPLC仪器制造商均以此产品作为仪器的进样器。工作原理:1、手柄位进样(Load)位置时,样品经微量进样针从进样孔注射进定量环,定量环充满后,多余样品从放空孔排出;2、将手柄转动至进样(Inject)位置时,阀与液相流路接通,由泵输送的流动相冲洗定量环,推动样品进入液相分析柱进行分析。虽然六通阀进样器具有结构简单、使用方便、寿命长、日常无需维修等特点,但正确的使用和维护将能增加使用寿命,保护周边设备,同时增加分析准确度。如使用得当的话,六通阀进样器一般可连续进样3万次而无需维修。以下浅谈有关六通阀进样器的使用及保养事宜(仅供参考):1、手柄处于Load和Inject之间时,由于暂时堵住了流路,流路中压力骤增,再转到进样位,过高的压力在柱头上引起损坏,所以应尽快转动阀,不能停留在中途。在HPLC系统中使用的注射器针头有别于气相色谱,是平头注射器。一方面,针头外侧紧贴进样器密封管内侧,密封性能好,不漏液,不引入空气;另一方面,也防止了针头刺坏密封组件及定子。2、六通阀进样器的进样方式有部分装液法和完全装液法两种。使用部分装液法进样时,进样量最多为定量环体积的75%,如20gL的定量环最多进样15p,L的样品,并且要求每次进样体积准确、相同;使用完全装液法进样时,进样量最少为定量环体积的3至5倍,即20gL的定量环最少进样60至1001aL的样品,这样才能完全置换样品定量环内残留的溶液,达到所要求的精密度及重现性。推荐采用100ul的平头进样针配合20ul满环进样。3、可根据进样体积的需要自已制作定量环,一般不要求精确计算定量环的体积,譬如,一根名义上10gL的定量环,实际是9gL还是1lgL并不重要,因为被测样品和校正样品的进样体积保持一致,在计算结果时误差都被抵消了。4、进样样品要求无微粒和能阻死针头及进样阀的物质,样品溶液均要用0.45~tm的滤膜过滤。防止微粒阻塞进样阀和减少对进样阀的磨损。为防止缓冲盐和其它残留物质留在进样系统中,每次结束后应冲洗进样器,通常用不含盐的稀释剂、水或不含盐的流动相冲洗,在进样阀的Load和Inject位置反复冲洗,再用无纤维纸擦净注射器针头的外侧。
