免疫等电聚焦分子式:CAS号:性质: 又称免疫电聚焦。是电泳分析方法之一。实际上它是免疫扩散电泳和等电聚焦技术相联合的一种电泳分析方法。具体操作大致有三种情况。(1)圆盘免疫等电聚焦是用聚丙烯酰胺凝胶柱先行等电聚焦,把微克量样品(如混合蛋白质)分开之后,把这凝胶柱条再包埋在具有缓冲体系的琼脂凝胶中,各蛋白随即扩散到琼脂胶内,并与抗血清槽(在琼脂胶板上)内扩散开的抗血甭(抗体)结合而成沉淀线;(2)把上述已分离的蛋白(在聚丙烯酰胺凝胶中)发刀下来填补在琼脂凝胶板的孔中,用双扩散试验法分析其中的特异性抗原;(3)像免疫电泳那样直接在玻片上作琼脂免疫等电聚焦。这一方法主要用于鉴定抗原。以免疫电泳优越之处在于能在选定的PH范围内完成蛋白(抗原)的分离,并提高了分辨力。
