地西泮【通用名称】地西泮【其他名称】地西泮 地西泮 拼音名：Dixipan 英文名：Diazepam 书页号：2000年版二部－214 C16H13ClN2O 284.74 本品为1-甲基-5- 苯基-7- 氯-1，3-二氢-2H-1,4-苯并二氮杂{卓} -2-酮。按干燥品 计算，含C16H13ClN2O 不得少于98.5％。 【性状】 本品为白色或类白色的结晶性粉末；无臭，味微苦。 本品在丙酮或氯仿中易溶，在乙醇中溶解，在水中几乎不溶。 熔点 本品的熔点（附录Ⅵ C）为130 ～134 ℃。 吸收系数 取本品，精密称定，加0.5 ％硫酸的甲醇溶液溶解并定量稀释使成每 1 ml中约含10μg 的溶液，照分光光度法（附录Ⅳ A），在284nm 的波长处测定吸收度， 吸收系数（Ｅ1% 1cm）为440～468 。 【鉴别】 (1) 取本品约10mg，加硫酸3ml 振摇使溶解，在紫外光灯(365nm) 下检 视，显黄绿色荧光。 (2) 取本品20mg，用氧瓶燃烧法（附录Ⅶ C）进行有机破坏，以5％氢氧化钠溶液 5ml 为吸收液，燃烧完全后，用稀硝酸酸化，并缓缓煮沸2 分钟，溶液显氯化物的鉴别 反应（附录Ⅲ）。 (3) 取本品，加0.5 ％硫酸的甲醇溶液制成每1ml 中含5μg的溶液，照分光光度法 （附录Ⅳ A）测定，在242nm、284nm与366nm 的波长处有最大吸收；在242nm波长处的吸 收度约为0.51，在284nm 波长处的吸收度约为0.23。 (4) 本品的红外光吸收图谱应与对照的图谱（光谱集138 图）一致。 【检查】 溶液的澄清度与颜色 取本品0.1g，加乙醇20ml,振摇使溶解，溶液应澄 清无色；如显色，与黄色1 号标准比色液（附录Ⅸ A第一法）比较，不得更深。 氯化物 取本品1.0g，加水50ml，振摇10分钟，滤过，分取滤液25ml，依法检查（ 附录Ⅷ A）与标准氯化钠溶液7.0ml 制成的对照液比较，不得更浓(0.014％) 。 有关物质 取本品，加丙酮制成每1ml 中含100mg 的溶液，作为供试品溶液；精密 量取适量，加丙酮稀释成每1ml 中含0.3mg 的溶液，作为对照溶液。照薄层色谱法（附 录Ⅴ B）试验，吸取上述两种溶液各5μl，分别点于同一硅胶ＧＦ254 薄层板上，以醋 酸乙酯－己烷(1:1) 为展开剂，展开后，晾干，置紫外光灯(254nm) 下检视。供试品溶 液如显杂质斑点，与对照溶液的主斑点比较，不得更深。 干燥失重 取本品，在105 ℃干燥至恒重，减失重量不得过0.5 ％（附录Ⅷ L）。 炽灼残渣 不得过0.1 ％（附录Ⅷ N）。 【含量测定】 取本品约0.2g，精密称定，加冰醋酸与醋酐各10ml使溶解，加结晶 紫指示液1 滴，用高氯酸滴定液(0.1mol/L)滴定，至溶液显绿色。每1ml高氯酸滴定 液(0.1mol/L)相当于28.47mg 的C16H13ClN2O 。 【类别】 抗焦虑药、抗惊厥药。 【贮藏】 密封保存。 【制剂】 (1) 地西泮片 (2) 地西泮注射液
