旋转酶分子式:CAS号:性质:又称回旋酶,促旋酶,旋转酶。属于解链酶(unwinding enzymes)类中的一种,属拓扑异构酶II(type II topoisomerase;topo II),该酶首先在大肠杆:菌中发现。分别由两个α亚基和两个β亚基组成,分子量4×105(其中,α亚基分子量约1.05×105,β亚基分子量约9.5×104)。它的作用是在水解ATP的同时能使松弛态环状DNA转变为负超螺旋DNA。这一作用很复杂,涉及三个步骤:(1)首先DNA回旋酶与DNA结合,使环状DNA扭曲而形成一个“右手结”结构,在这一过程中形成一个稳定的正超螺旋,同时又引入一个负超螺旋;(2)然后该酶在右手结的背后打断双链DNA,并将其搭在另一条双链的前面,这样就将右手性正超螺旋变为左手性负超螺旋;(3)最后将断点连接起来。DNA负超螺旋的引入能使打断碱基对所需的能量降低约4.1kJ mol-1,有利于将DNA双链分开。在复制过程中,当DNA新链在模板上形成后,DNA回旋酶将复制好的DNA双链变为天然的负超螺旋的构型,此过程需ATP水解供能。若缺乏ATP,则该酶催化负超螺旋松弛。
