亲和色谱分子式:CAS号:性质:又称亲和色谱。利用生物大分子所具有的专一亲和力而设计的一种层析技术。用连接在基体的配位体(如酶、抗体、激素等)作为固定相,使与蛋白质或其他大分子发生可逆的高选择性的相互作用,利用不同亲和力进行分离的液相色谱法。(1)酶-底物或底物类似物或抑制剂或辅因子(辅酶、金属离子等);(2)抗体-抗原或病毒或细胞;(3)激素、维生素-受体蛋白或载体蛋白;(4)外源凝集素-多糖化合物或糖蛋白或细胞表面受体蛋白或细胞;(5)核酸-互被碱基链段或组蛋白或核酸聚合酶或核酸结合蛋白。层析柱的制备是将配体共价偶联到一个不溶性的基物上。样品溶液通过此层析柱时,与此配体能特异结合的组分便被吸附而与其他组分分开。然后,在不能发生特异结合的条件下用某些试剂淋洗以将该物质与配体重新拆开,这样便可获得纯化后的该组分。亲和层析法具有简便、快速、高效且分离产物不会失活等优点。其主要用途是分离和提纯蛋白质、膜和多糖类物质。
