1.In order to search for more potent and less toxic antithrombotic agents, eleven derivatives of 6-(4-substituted-phenyl)-4, 5-dihydro~3(2H)-pyridazinones were designedand according to mechanism of action and structure-activity relationship of dihydropyridazinone compounds.为了寻找优良的血小板聚集抑制剂,本课题依据二氢哒嗪酮类化合物的作用机理和构效关系,在保留二氢哒嗪酮类化合物基本结构的基础上适当延长碳链,引入各种不同的侧链及杂环,共设计并合成了十一个二氢哒嗪酮类化合物,均为首次报道。
2.In order to search for more potent and less toxic antithrombotic agents twenty-eight derivatives of 6-(4-substituted aminophenyl)-5-methyl-4,5-dihydro-3(2H)-pyridazinones and 6-(4- substituted aminophenyl)-4,5-dihydro-3(2H)-pyridazinoneswere designed and synthesized according to mechanism of action and structure-activity relationship of dihydropyridazinone compounds.二氢哒嗪酮类化合物具有良好的心血管药理作用如抗血小板凝集和强心等,有的已成为药物如匹莫苯(pimobendan),有的已结束Ⅲ期临床研究如MCI-154,有些正在进行临床前研究如SMⅡ_4、Y909、DMDP和CCI-17810等,这类化合物在心脑血管疾病的治疗方面具有广阔的应用前景。

二酮哌嗪分子式:CAS号:性质:按氧基在哌嗪环上位置的不同,有2,3-,2,5-,3,5-二酮哌嗪等异构体。2,3-二酮哌嗪,熔点285℃,由乙二胺和草酸作用可制得。2,5-二酮哌嗪,片状结晶,熔点318~320℃(分解),溶于乙醇及水。其衍生物是相当重要的一类化合物,可看成是α-氨基酸的脱水环状二聚物,典型代表物有2,5-二酮-3,3,6,6-四甲基哌嗪(没有熔点,在360℃左右升华)。3,5-二酮哌嗪,在210℃左右分解,由亚氨二醋酸与脲素共热可制得。均用于有机合成。
