胞壁质酶分子式：暂无分子量：14400±100CAS号：9001-63-2性质：蛋白质溶菌酶是由129个氨基酸组成的均一链的多肽。为白色冷冻干粉或结晶。等电点为10.5-11.0。酸性溶液较为稳定，加热到55℃无影响，能溶于水。制备方法：从蛋清中提取制得。1.原料处理 新鲜或冰冻蛋清（或蛋壳水）70kg，用试纸测其pH值，应在8.0左右，经解冻后通过铅筛，除去蛋清中的脐带、蛋壳碎片及其他杂质。2.吸附 用冰水冷却并搅拌蛋清，使温度降到5℃，缓缓加入724树脂10kg，搅拌，使树脂完全悬浮在蛋清中。保持温度0-10℃，再继续搅拌5h。将蛋清移入冷库，在0-10℃静置过液。3.洗涤、洗脱 隔天把上层蛋清清液倾出，下层用清水洗去附着的蛋白质，先后共洗四次。最后将树脂抽滤去水分，用24kg0.15M磷酸缓冲液（pH6.5）分三次加入树脂搅拌，每次搅拌15min后抽滤去水分。然后用10%硫酸铵18kg，分4次洗脱溶菌酶。合并洗脱液，加入32%（重量/体积）固体硫酸铵使含量达40%，析出白色沉淀。在冷处放置过夜，过滤出沉淀。4.透析 将沉淀溶解于1.5L蒸馏水中，在冷库内透析24-36h，除去大部分硫酸铵。5.盐析 透析完毕，将溶液用滤纸过滤，得澄清透析液。用滴管慢慢加入氢氧化钠溶液，使pH上升到8.5-9.0，如有白色沉淀，应即离心除去。然后在搅拌下加入3N盐酸，使溶液pH为3.5。按体积缓缓加入5%固体氯化钠，生成白色沉淀。在0-10℃冷库放置48h，抽滤得到溶菌酶沉淀。6.干燥沉淀内加入10倍量0℃无水丙酮，搅拌使颗粒松细，在冷处静置数小时，滤去丙酮。沉淀用五氧化二磷真空干燥，即得口服或外用溶菌酶粉剂原料。如不用丙酮脱水，可将溶菌酶透析液冷冻干燥可得不含氯化钠的溶菌酶成品。按蛋清重量计，收率0.1%。用途：主要用于五管科多种粘膜疾患，如副鼻窦炎、慢性鼻炎、咽喉炎、扁平苔癣、口腔溃疡、渗出性中耳炎等徉患，亦用于皮肤科带状包疹，扁平疣及铬性鼻炎。
