青霉素酶分子式:CAS号:性质:催化水解青霉素β-内酰环产生青霉噻唑酸的一类β-内酰胺酶。国际生物化学联合会(IUB)酶学委员会的酶编号为3.5.2.6。在各种微生物中分布广泛,特别在细菌中更为广泛。由蜡状芽孢杆菌Bacillus cereus 5/B与B. cereus 569分别得到分子量为35200与31500的青霉素酶结晶,从巨大芽孢杆菌B. megaterium 提取出α,β,γ青霉素酶结晶,随后从不同细菌又分离出多种青霉素酶。青霉素酶对热极不稳定,分解β-内酰胺的速度因青霉素与酶的种类不同而异,但对6-氨基青霉烷酸(6-APA)作用却很微弱。酶反应最适PH值为6.0~6.5. 以青霉素G钾盐为底物,Km为4.32×10-3M(5/B酶);4.89×10-3M(569酶)。细菌产生青霉素酶导致出现耐药性,临床上已分离出多种类型青霉素酶,白色粉末。分子量50000。溶于水。酶溶液易失活,冻干品2~8℃可稳定一周。青霉素酶作用于青霉素的β-内酰胺环,使青霉素转变为无抗菌活性的青霉素酮酸(penicilloic acid)。医疗上用于青霉素过敏症状的治疗。青霉素酶在实验中用于破坏或抑制青霉素活性。
