柱后衍生化法分子式:CAS号:性质:将多组分样品先注入色谱柱,按选定的色谱条件使之在色谱柱上分离,当各个组分从色谱柱流出后,在一定的反应条件下分别与衍生化试剂反应,生成的衍生化产物再进入检测器检测。常用的衍生化方法有紫外衍生、荧光衍生、电化学衍生和手性衍生化法等。柱后衍生化法的最大优点是不改变被分离组分的色谱行为。在柱后衍生化过程中要注意流动相组成与反应介质的一致性,必须考虑衍生化试剂、反应产物的溶解度,以及它们与流动相可能发生的副反应;同时要求被测组分从色谱柱出来后到达检测器之间的体积要非常小,尽量减少组分色谱带变宽而影响分离;而且要选择反应速度快的衍生化试剂,使衍生化反应在短时间内进行完全。
