丙硫氧嘧啶【通用名称】丙硫氧嘧啶【其他名称】丙硫氧嘧啶 丙硫氧嘧啶 拼音名:Bingliuyang Miding 英文名:Propylthiouracil 书页号:2000年版二部-99 C7H10N2OS 170.24 本品为6-丙基-2-硫代-2,3-二氢-4(1H)嘧啶酮。按干燥品计算,含C7H10N2OS不 得少于98.0%。 【性状】 本品为白色结晶或结晶性粉末;无臭,味苦。 本品在乙醇中略溶,在水中极微溶解,在氢氧化钠试液或氨试液中溶解。 熔点 本品的熔点(附录Ⅵ C)为218~221℃。 【鉴别】 (1) 取本品的饱和水溶液,加热至沸,加入等量新配制的含0.4%亚硝 基铁氰化钠、0.4 %盐酸羟胺与0.8%的碳酸钠的混合溶液,即显绿蓝色。 (2) 取本品约25mg,滴加溴试液至完全溶解,加热,退色后,放冷,滴加氢氧化钡 试液,即生成白色沉淀。 (3) 本品的红外光吸收图谱应与对照的图谱(光谱集70图)一致。 【检查】 硫脲 取本品0.50g ,加水50ml;加热回流使溶解,放冷,滤过,取滤 液5.0ml,加水稀释至50ml,摇匀,取10ml置纳氏比色管甲中;加0.010%硫脲溶液1.0ml, 另取上述滤液10ml与水1.0ml ,置纳氏比色管乙中;在甲乙两管中分别加入醋酸钠0.50g 与硝酸银试液5.0ml后,置水浴中加热5分钟,乙管所显颜色与甲管比较,不得更深。 干燥失重 取本品,在105℃干燥至恒重,减失重量不得过0.5%(附录Ⅷ L)。 炽灼残渣 取本品1.0g,依法检查(附录Ⅷ N),遗留残渣不得过0.1%。 重金属 取炽灼残渣项下遗留的残渣,依法检查(附录Ⅷ H第二法),含重金属不 得过百万分之二十。 【含量测定】 取本品约0.3g,精密称定,加氢氧化钠试液5ml与水50ml,必要时 加热使溶解,放冷至室温,加酚酞指示液2滴,滴加稀醋酸至红色消失,加二苯偕肼指 示液1ml ,用硝酸汞滴定液(0.05mol/L)滴定,至溶液显淡紫堇色。每1ml硝酸汞滴定液 (0.05mol/L) 相当于17.02mg的C7H10N2OS 。 【类别】 抗甲状腺药。 【贮藏】 遮光,密封保存。 【制剂】 丙硫氧嘧啶片
