磷霉素钠【通用名称】磷霉素钠【其他名称】磷霉素钠 磷霉素钠 拼音名:Linmeisuna 英文名:Fosfomvcin Sodium 书页号:2000年版二部-1076 C3H5Na2O4P 182.02 本品为(-)(1R,2S)-1,2-环氧丙基磷酸二钠盐。按无水物计算,每1mg的效价不得少于700磷霉 素单位。 【性状】本品为白色结晶性粉末;无臭,味咸;在空气中极易潮解。 本品在水中易溶,在甲醇中微溶,在乙醇或乙醚中几乎不溶。 比旋度 取本品,精密称定,加水制成每1ml中含50mg的溶液,依法测定(附录Ⅵ E),比旋度为 -4.2°至-5.5°。 【鉴别】(1) 取本品及磷霉素标准品,分别加0.2mol/L乙二胺四醋酸二钠溶液制成每1ml中含 20mg的溶液。照薄层色谱法(附录Ⅴ B)试验,吸取上述两种溶液各2μl,分别点于同一硅胶G薄层板 上,以异丙醇-醋酸乙酯-水(4:2:4)为展开剂,展开后,晾干,喷以显色液(取磷钼酸5g,加醋酸 100ml,再加硫酸5ml),置120℃加热15分钟后检视。供试品溶液所显主斑点的颜色和位置应与标准品 溶液的主斑点相同。 (2)本品显钠盐的火焰反应(附录Ⅲ)。 【检查】碱度 取本品,加水制成每1ml中含50mg的溶液,依法测定(附录Ⅵ H),pH值应为9.0~ 10.5。 溶液的澄清度与颜色 取本品5份,各1.1g,分别加水5ml溶解,溶液应澄清无色;如显浑浊,与 1号浊度标准液(附录Ⅸ B)比较,均不得更浓;如显色,与黄色1号标准比色液(附录Ⅸ A 第一法)比 较,均不得更深。 二醇物 取本品约0.2g,精密称定,置250ml碘瓶中,加水100ml与邻苯二甲酸氢钾缓冲液 (pH6.4)50ml,精密加入0.005mol/L高碘酸钠溶液10ml,摇匀,密塞,于25℃避光放置90分钟,加入 10%碘化钾溶液10ml,密塞,避光放置2分钟后,用硫代硫酸钠滴定液(0.01mol/L)滴定,午近终点 时,加淀粉指示液1ml,继续滴定至蓝色消失,并将滴定的结果用空白试验校正。每1ml硫代硫酸钠滴 定液(0.01mol/L)相当于0.5001mg的C3H7Na2O5P,含二醇物不得过0.5%。 磷含量 取本品约0.1g,精密称定,加0.05mol/L高碘酸钠溶液40ml及高氯酸2ml,水浴中加热1 小时,放冷,转移至200ml量瓶中,加水溶解并稀释至刻度,摇匀,精密量取10ml,置100ml量瓶中, 加碘化钾试液1ml,溶液显棕色,加硫代硫酸钠试液至溶液无色为止,再加水至刻度,摇匀,作为供试 品溶液;另取磷酸二氢钾约70mg,精密称定,与供试品溶液同法制备磷对照溶液,并按同法制成空白 溶液。精密量取上述三种溶液各5ml,分别置25ml量瓶中,各加钼酸铵试液2.5ml和1-氨基-2-萘酚-4- 磺酸试液1ml,分别加水稀释至刻度,摇匀,于20℃±1℃放置30分钟,照分光光度法(附录Ⅳ B),在 740nm的波长处,用水做参比,测定吸收度。按下式计算磷含量,磷含量应为16.2%~17.9%。 A1-A3 W2 磷含量(%)=————×——×22.76 A2-A3 W1 式中 A1为供试品溶液的吸收度; A2为磷对照溶液的吸收度; A3为空白溶液的吸收度; W1为供试品的重量; W2为磷酸二氢钾的重量; 22.76为磷酸二氢钾中磷的含量(%)。 水分 取本品,照水分测定法(附录Ⅷ M 第一法 A)测定,含水分不得过1.5%。 重金属 取本品1.0g,依法检查(附录Ⅷ H 第三法),含重金属不得过百万分之十。 热原 取本品,加灭菌注射用水制成每1ml中含7万单位的溶液,依法检查(附录Ⅺ D),剂量按家 兔体重每1kg注射1ml,应符合规定。 无菌 取本品,分别加入100ml0.9%无菌氯化钠溶液中使溶解,用薄膜过滤法处理后,依法检查 (附录Ⅺ H),应符合规定。 【含量测定】精密称取本品适量,加灭菌水制成每1ml中约含500单位的溶液,照抗生素微生物检 定法(附录Ⅺ A)测定,1000磷霉素单位相当于1mg的C3H7O4P。 【类别】抗生素类药。 【贮藏】严封,在凉暗干燥处保存。 【制剂】注射用磷霉素钠
