本发明属于生物技术制药领域,具体涉及长效纤连蛋白iii型结构域融合蛋白及其制备与应用。

自1982年世界上第一个重组蛋白药物-重组人胰岛素上市以来,蛋白和多肽类药物现已发展成为现代生物制药领域最重要的品类之一。目前fda已经批准180多种蛋白和多肽药物用于临床。

与传统的小分子化学药物相比,蛋白和多肽类药物具有特异性强、毒性低、副作用小、生物功能明确等优势,在某些疾病(如糖尿病、血友病、蛋白酶缺少导致的罕见病等)的治疗中具有不可替代的作用[2015重组蛋白药物研发及专利分析]。除了抗体及fc融合的蛋白,大多数蛋白和多肽类药物的分子量通常都小于肾小球滤过消除的上限(60kd),同时它们易被机体的蛋白酶或肽酶代谢消除,因此这类药物的血浆半衰期往往很短。如天然胰高血糖素样肽-1(glucagon-likepeptide1,glp-1)易被二肽肽酶4(dipeptidylpeptidase4,dpp-4)酶切降解,其血浆半衰期仅有1-2min。此外,几乎所有的蛋白和多肽类药物都是通过非肠道途径给药,为了达到治疗效果,须频繁或高剂量给药,病人的依从性低。因此,旨在降低蛋白和多肽药物对蛋白酶的敏感性、延长其血浆半衰期的蛋白修饰技术已被用于改善这些药物的药代动力学过程。

药物在体内的药代动力学过程包括吸收、分布、代谢和消除。蛋白质和多肽药物由淋巴系统吸收,广泛分布于中心腔体的细胞外空间,经由蛋白酶和肽酶酶解代谢后,大分子蛋白通过受体介导的机制被机体消除,而分子量小于60kd的蛋白和多肽更易被肾小球滤过消除。对于小分子蛋白和多肽来说,目前临床上采用的延长其血浆半衰期的策略可分为2大类:1)将蛋白或多肽与天然长半衰期的蛋白或蛋白结构域如fc、人血白蛋白(humanserumalbumin,hsa)等融合,通过fcr介导的机制促进蛋白在体内再循环;2)将蛋白或多肽与惰性多肽如聚乙二醇(poly-ethyleneglycol,peg)、xten(也称重组peg,rpeg)、等融合,增加蛋白的表观分子量,避免其被肾小球滤过消除。

在血浆蛋白中,igg、hsa和转铁蛋白的半衰期要长于其他蛋白。这3种蛋白相对分子质量均大于肾脏过滤阈值,igg和hsa还能通过新生儿fc受体(neonatalfcreceptor,fcrn)介导的机制在体内再循环,而转铁蛋白则通过网格蛋白依赖的转铁蛋白受体延长体内停留的时间,因此它们都有较长的体内半衰期。蛋白质、多肽和这些蛋白融合后,往往可以显著增加半衰期。

如上所述,人免疫球蛋白igg1,igg2,igg4亚型可通过依赖于ph的fcrn介导在体内进行再循环,因此这些igg亚型的血浆半衰期可达3-4周。免疫球蛋白与fcrn的作用位点为fc区域,在酸性环境下,igg与细胞膜上的fcrn相结合,避免了溶酶体的降解。在中性环境下,重新被释放到血液中。fc融合是目前研究最多、进展最快的蛋白融合技术。fc融合蛋白不仅可以提高半衰期、保留融合蛋白的生物活性,还具有fc的抗体活性。此外,fc融合技术还具有其他优点-----与fcrn受体的结合为融合蛋白提供了新的体内吸收方式,如epo、促卵泡激素和干扰素-α/β与fc融合后能通过内皮细胞,因此可通过上呼吸道吸入给药[kuott,bakerk,yoshidam,etal.neonatalfcreceptor:fromimmunitytotherapeutics.jclinimmunol2010;30(6):777-789]。自第一个fc融合的药物enbrel(etanercept)于1998被fda批准上市至2015年5月,已有11个fc融合的重组蛋白批准用于临床。这些fc融合的重组蛋白的血浆半衰期均比未融合的蛋白大大延长,如fc融合的因子ix(alprolix,eftrenonacog-alpha;大约98kd的因子ix-fc)血浆半衰期为57-83h,是单独因子ix(半衰期18h)的3倍多[shapiroad,ragnimv,valentinola,etal.recombinantfactorix-fcfusionprotein(rfix-fc)demonstratessafetyandprolongedactivityinaphase1/2astudyinhemophiliabpatients.blood2012;119:666-72][powelljs,pasikj,ragnimv,etal.phase3studyofrecombinantfactorixfcfusionproteininhemophiliab.nengljmed2013;369:2313-23]。天然glp-1的体内半衰期仅2min,美国礼来公司开发的trulicity(dulaglutide;glp-1-fc融合蛋白)血浆半衰期达4-5天,临床使用中仅需每周给药一次,大大提高了患者用药的依从性,该药2014年被批准上市[glaesnerw,vickam,millicanr,etal.engineeringandcharacterizationofthelong-actingglucagon-likepeptide-1anagloguely2189265,anfcfusionprotein.diabetesmetabresrev.2010;26:287–96.]。目前,fc主要是与受体膜外区以及多肽形成融合蛋白,其它类型fc融合蛋白进入临床后期开发的较少;由于结构限制,fc一般融合在靶标蛋白的c端并形成二聚体,这种融合方式由于构象干扰和空间位阻,往往导致蛋白稳定性降低、生物学活性显著减弱。此外,fc主要采用igg1亚型的fc,可能会有由于adcc和cdc活性带来的不必要的副作用。

hsa是血浆中含量最高的蛋白,在血浆ph维持、代谢物和脂肪酸转运及稳定血压中发挥重要作用。与igg类似,hsa可通过依赖于ph的fcrn介导在体内进行再循环,分子量(mw=66.5kd)刚好超过肾小球滤过消除的上限,且带强负电荷,会被肾小球基底膜排斥,因此其血浆半衰期长达19天。由于其水溶性良好、无免疫原性、组织分布广、无酶活、且易于在肿瘤和炎症组织部分聚集等特性,将hsa作为融合伴侣,将其连接到半衰期较短的蛋白类药物上,不仅可以增加药物分子的相对分子质量和水化半径,而且可以利用fcrn介导的再循环机制延长药物的半衰期。第一个hsa融合的药物是gsk公司2014年上市的tanzeum(glp1-hsa),hsa的融合将天然glp-1的半衰期从1-2天延长至4-7天。与礼来公司的trulicity类似,tanzeum每周仅需给药一次,大大提高了患者用药的依从性。conjuchemllc公司的pc-dactm技术,在给药前多肽药物与hsa相结合,使glp-1半衰期延长了6000倍以上[bossed,prausm,kiesslingp,etal.phaseicomparabilityofrecombinanthumanalbuminandhumanserumalbumin.jclinpharmacol2005,45(1):57-67]。teva公司的balugrastim(gcsf-hsa),目前已完成临床ⅲ期试验,其半衰期比单独的g-csf提高了7倍以上。csl公司的csl654(rfix-fp),csl689(rfviia-fp),诺华公司的albuferon(ifn-a2b-has),merrimack公司的mm-111(her3-hsa-her2双特异性抗体融合蛋白)等均在临床试验阶段。与hsa融合的蛋白药物开发过程中也发现一些问题,如目的蛋白与hsa的融合可能会导致目的蛋白活性有所下降、药用价值降低,融合蛋白在发酵、纯化和储存过程中还易发生降解和聚合现象等问题。

peg是一种高度柔性、不带电荷、几无免疫原性的亲水性聚合物,其被fda认定为gras(generallyrecognizedassafe),并批准用于延长蛋白或多肽药物的半衰期已经有20多年的历史。目前已有12个peg化的药物被fda成功应用于临床,如治疗乙型肝炎的(pegylatedinterferonalfa-2b)、(pegylatedifn-a2a)、治疗化疗诱导的噬中性粒细胞减少症的(pegfilgrastim,peg缀合的粒细胞集落刺激因子)、治疗慢性肾病病人贫血症的mycera(红细胞生成素-b的一种peg化形式)等[turecekpl,bossardmj,schoetensf,ivensia.pegylationofbiopharmaceuticals:areviewofchemistryandnonclinicalsafetyinformationofapproveddrugs.jpharmsci.2016feb;105(2):460-75]。peg修饰增加了蛋白的水溶性和表观分子量,减少肾脏对蛋白质的滤过消除,保护蛋白质免遭酶解,因此其给药频率可减少至一周一次。由于peg的多分散性和异质性,peg修饰的蛋白药物,需要在蛋白质上进一步修饰,因此对于质量研究、工艺控制、产品质控都有很高的挑战。对于分子质量相对小的细胞因子、生长激素等蛋白来说,peg化带来的位阻效应阻碍与相应受体的结合而导致表观活性降低。peg在生物体内不可降解,当长期大剂量注射peg-干扰素(peg-ifna2a)后peg会在肾脏中积累(conovercdetal.artificialorgans197;21:36-378;bendeleaetal.toxicolsci1998;42:152-157);非临床毒性研究显示,fda批准的12个peg修饰的药物中有5个药物可导致细胞空泡的形成,且空泡的形成与peg有关,其中2个()与多个分子量小的peg偶联(分别为5kd和10kd),另外3个()与分子量40kd的单一peg分子偶联[ivensia,achanzarw,baumanna,etal.pegylatedbiopharmaceuticals:currentexperienceandconsiderationsfornonclinicaldevelopment.toxicolpathol.2015;43(7):959-983]。此外,peg价格不菲,需和蛋白进行化学偶联以及后续的纯化等,因此从药物设计的角度来说peg的应用仍受到很大的限制[feecj,vanalstinejn.purificationofpegylatedproteins.in:jansonj-c,editor.proteinpurification:principles,highresolutionmethods,andapplications.3rded.newyork:wiley;2011.p.339-62.]。

peg模拟物xten是由amunix公司开发的由6种亲水、结构稳定的氨基酸(丙氨酸ala、谷氨酸glu、甘氨酸gly、脯氨酸pro、丝氨酸ser、苏氨酸thr)组成的可体内降解、无免疫原性的不定型多聚体。研究显示在不同的动物模型(小鼠、大鼠、猴子)中,288aa(32kd)-1008aa(111kd)的xten融合可延长艾塞那肽的半衰期从50-125倍不等[schellenbergerv,wangcw,geethingnc,etal.arecombinantpolypeptideextendstheinvivohalf-lifeofpeptidesandproteinsinatunablemanner.natbiotechnol.2009;27:1186–90]。xten化艾塞那肽(vrs-859)目前已进入临床i期,用于2型糖尿病人的血糖控制;vrs-859在小鼠和大鼠模型上的半衰期比艾塞那肽延长65-71倍,在猴子身上则从30min延长至60h[schellenbergerv,wangcw,geethingnc,etal.arecombinantpolypeptideextendstheinvivohalf-lifeofpeptidesandproteinsinatunablemanner.natbiotechnol.2009;27:1186–90]。另一个进入临床iii期的xten融合蛋白是vrs-317(xten化的hgh),研究结果显示vrs-317的人体半衰期可达131h[yuenkcj,conwaygs,popovicv,etal.along-actinghumangrowthhormonewithdelayedclearance(vrs-317):resultsofadouble-blind,placebo-controlled,singleascendingdosestudyingrowthhormone-deficientadults.jclinendrocrinmetab.2013;98:2595–603.],远高于peg化rhgh及ctp-hgh融合蛋白mod-4023[strohlwr.fusionproteinsforhalf-lifeextensionofbiologicsasastrategytomakebiobetters.biodrugs.2015aug;29(4):215-39]。与peg类似,对于分子质量相对小的细胞因子、生长激素等蛋白来说,xten化带来的位阻效应会导致蛋白的生物活性降低;另外,xten属于高水溶性聚合物,往往会导致融合的蛋白粘度变大,分离和提纯比较费力,做成制剂比较困难,且目前还未有正式批准用于临床的融合蛋白,因此,其安全性有待进一步评估。

然而,本技术领域中还需要开发其他能够用来与蛋白质或肽类药物生成融合蛋白的融合伴侣分子。

发明概述

本发明人发现,纤连蛋白iii型结构域是一种优秀的融合伴侣分子,将其与多种蛋白质或肽融合后,可以显著增加该蛋白质或肽的体内半衰期。由此完成了本发明。

具体地,本申请提供下述实施方案:

1.纤连蛋白iii型结构域融合蛋白,其包含如下部分:

纤连蛋白iii型结构域;

一个或多个连接肽;和

第一生理活性肽。

2.实施方案1所述的纤连蛋白iii型结构域融合蛋白,其中,第一生理活性肽插入在所述纤连蛋白iii型结构域的内部。

3.实施方案2所述的纤连蛋白iii型结构域融合蛋白,其中,第一生理活性肽插入在所述纤连蛋白iii型结构域的相邻2条β链之间形成的柔性loop环内,例如选自下组的环内:ab环、bc环、cd环、de环、ef环或fg环;优选地,第一生理活性肽插入所述纤连蛋白iii型结构域的cd环或fg环内。

4.实施方案1所述的纤连蛋白iii型结构域融合蛋白,其中,第一生理活性肽通过连接肽连接到纤连蛋白iii型结构域的n端或c端。

5.实施方案1-4中任一项所述的纤连蛋白iii型结构域融合蛋白,其还包含第二生理活性肽。

6.实施方案5所述的纤连蛋白iii型结构域融合蛋白,其中第二生理活性肽插入在所述纤连蛋白iii型结构域的内部,条件是当第一生理活性肽亦插入在所述纤连蛋白iii型结构域的内部时,第二生理活性肽和第一生理活性肽插入在纤连蛋白iii型结构域的不同位置。

7.实施方案5所述的纤连蛋白iii型结构域融合蛋白,其中第二生理活性肽通过连接肽连接到纤连蛋白iii型结构域的n端或c端,条件是当第一生理活性肽亦连接到纤连蛋白iii型结构域的n端或c端时,第二生理活性肽和第一生理活性肽连接到纤连蛋白iii型结构域的相反末端。

8.实施方案7所述的纤连蛋白iii型结构域融合蛋白,其中,第二生理活性肽插入在所述纤连蛋白iii型结构域的相邻2条β链之间形成的柔性loop环内,例如选自下组的环内:ab环、bc环、cd环、de环、ef环或fg环。

9.实施方案1-8中任一项所述的纤连蛋白iii型结构域融合蛋白,其中,纤连蛋白iii型结构域为纤连蛋白第7iii型结构域(fn7),尤其是序列如seqidno:2所示的fn7。

10.实施方案1-8中任一项所述的纤连蛋白iii型结构域融合蛋白,其中,纤连蛋白iii型结构域为纤连蛋白第10iii型结构域(fn10),尤其是序列如seqidno:4所示的fn10。

11.实施方案1-10中任一项所述的纤连蛋白iii型结构域融合蛋白,其中该融合蛋白中的至少一个连接肽是柔性肽,即具有柔性结构的多肽。

12.实施方案11所述的纤连蛋白iii型结构域融合蛋白,其中该融合蛋白中的全部连接肽均为柔性肽。

13.实施方案11或12所述的纤连蛋白iii型结构域融合蛋白,其中,所述柔性肽由分子量小的极性氨基酸如甘氨酸(gly)、丝氨酸(ser)、苏氨酸(thr)、丙氨酸(ala)、谷氨酸(glu)或苯丙氨酸(phe)组成。

14.实施方案13所述的纤连蛋白iii型结构域融合蛋白,其中,所述柔性肽选自下组:(g4s)n,其中n=1、2、3、4或5;(gly)8,(gly)6,gggsggggs,ggggsgggs,gsagsaagsgef、kesgsvsseqlaqfrsld或egkssgsgseskst。

15.实施方案1-10中任一项所述的纤连蛋白iii型结构域融合蛋白,其中该融合蛋白中的至少一个连接肽是由α-螺旋组成的刚性肽。

16.实施方案15所述的纤连蛋白iii型结构域融合蛋白,其中该融合蛋白中的全部连接肽均为由α-螺旋组成的刚性肽。

17.实施方案15或16所述的纤连蛋白iii型结构域融合蛋白,其中,所述由α-螺旋组成的刚性肽的氨基酸序列选自下组:(eaaak)n,其中n=1、2、3、4或5;a(eaaak)na(n=2-5)。

18.实施方案1-17中任一项所述的纤连蛋白iii型结构域融合蛋白,其中,至少一个连接肽取代纤连蛋白iii型结构域的一个或多个氨基酸残基。

19.实施方案1-17中任一项所述的纤连蛋白iii型结构域融合蛋白,其中,连接肽不取代纤连蛋白iii型结构域n端或c端任何氨基酸残基。

23.实施方案5-22中任一项所述的纤连蛋白iii型结构域融合蛋白,其中第二生理活性肽选自下组:激素、细胞因子、酶、生长因子、转录调控因子、凝血因子、疫苗、结构蛋白质、配体蛋白质和受体、抗体或其抗原结合片段、毒性蛋白。

26.实施方案1-25中任一项所述的纤连蛋白iii型结构域融合蛋白,其还包含fc片段,其中该fc片段不是所述第一生理活性肽,亦不是所述第二生理活性肽。

27.实施方案26所述的纤连蛋白iii型结构域融合蛋白,具有如下的结构:

a—fn—fc—b

其中,a代表第一生理活性肽,b代表第二生理活性肽,—代表直接连接或者通过连接肽连接。

28.实施方案1所述的纤连蛋白iii型结构域融合蛋白,其包含如下任一项所示的氨基酸序列:seqidno:12、14、16、18、20、22、24、26、28、30、32、34、36、38、40、42、44、46、48、50、52、54、56、58、60、62。

29.一种多核苷酸,其编码如实施方案1-28任一项所述的纤连蛋白iii型结构域融合蛋白,优选地,所述多核苷酸包含选自下组的核苷酸序列:seqidno:11、13、15、17、19、21、23、25、27、29、31、33、35、37、39、41、43、45、47、49、51、53、55、57、59、61。

30.一种表达载体,其含有如实施方案29所述的多核苷酸。

31.一种宿主细胞,其包含如实施方案30所述的表达载体。

32.实施方案31所述的宿主细胞,其中该宿主细胞是用如实施方案29所述的表达载体瞬时转染的哺乳动物宿主细胞。

33.一种制备如实施方案1-28中任一项所述的纤连蛋白iii型结构域融合蛋白的方法,包括:在容许所述纤连蛋白iii型结构域融合蛋白表达的条件下培养实施方案32所述的哺乳动物宿主细胞;从培养上清中收集分泌的纤连蛋白iii型结构域融合蛋白。

34.实施方案1-28中任一项所述的纤连蛋白iii型结构域融合蛋白用于制备药物的用途。

35.纤连蛋白iii型结构域的用途,所述用途选自以下任一项或多项:1)提高生理活性肽的活性;2)提高生理活性肽的稳定性;3)延长生理活性肽的血浆半衰期。

36.实施方案35的用途,其中所述纤连蛋白iii型结构域为:

(a)纤连蛋白第7iii型结构域(fn7),尤其是氨基酸序列如seqidno:2所示的fn7;或

(b)纤连蛋白第10iii型结构域(fn10),尤其是氨基酸序列如seqidno:4所示的fn10。

37.实施方案34的用途,其中该用途是通过将纤连蛋白iii型结构域和生理活性肽制备成如实施方案1-22中任一项所述的融合蛋白而实现的。

38.实施方案34-37中任一项的用途,其中生理活性肽选自:激素、细胞因子、酶、生长因子、转录调控因子、凝血因子、疫苗、结构蛋白质、配体蛋白质和受体、抗体或其抗原结合片段、毒性蛋白;优选地,生理活性肽选自下组:人生长因子、集落刺激因子、fab重链、fab轻链、scfv。

本领域技术人员能够理解,当用给定的参考氨基酸或核苷酸序列描述某种蛋白质或核酸分子时,这样的蛋白质或核酸分子也涵盖与参考序列相比有一个、几个或多个氨基酸或核苷酸的变异,例如取代、缺失、插入或添加的序列,只要这样的变异不改变原始序列的功能。这样的变异可使得蛋白质或核酸分子的序列与给定的序列具有至少80%,至少85%,至少90%,至少95%,至少96%,至少97%,至少98%,至少99%,或至少99.5%的同源性,优选地,与参考序列相比的变异均存在于融合蛋白分子的生理活性肽部分之外。

本发明的有益效果

本发明的纤连蛋白iii型结构域融合蛋白,能够有效保护目标蛋白的n端和/或c端,使得目标蛋白不易被酶解,能够在体内保持其生理活性肽部分的生理活性的同时,显著增加生理活性肽的体内半衰期,和/或提高生理活性肽的体内稳定性。因此其体内半衰期更长,在临床上有望实现三周甚至一月一次的给药频率;所述融合蛋白的生产和制备工艺和抗体相似,产物表达量大。而且,本发明的融合蛋白不会产生在体内无法降解的副作用。

实施例

1.fn融合蛋白真核表达载体的构建

pcr扩增人gcsf(粒细胞集落刺激因子)、人gh(生长激素)、ucht1(抗-cd3抗体)fab重链(hc)、ucht1轻链(lc)、herceptin(抗her-2抗体)fab、herceptinlc、ucht1scfv、herceptinscfv、anti-cd19scfv基因(上述基因均由idt公司合成)。实施重叠pcr将人gcsf和人gh基因通过连接肽克隆至纤连蛋白(fibronectin,fn)第10结构域(fn10)或第7结构域(fn7)的loop环内,得到fn10-1a-gcsf、fn10-1g-gcsf、fn10-2a-gcsf、fn10-2g-gcsf、fn7-1g-gcsf和fn7-1g-hgh片段。将上述fn7-1g-gcsf和fn7-1g-hgh两个基因片段通过连接肽(gslinker或者a-helix)分别与人igg1fc的n端融合得到fn7-1g-gcsf-fc和fn7-1g-gh-fc。人igg1fc(ch2-ch2恒定区)含有7个突变(e233p,l234v,l235a,δg236,a327g,a330s和p331s)。实施重叠pcr将ucht1fabhc、ucht1lc、herceptinfabhc、herceptinlc、ucht1scfv、herceptinscfv、anti-cd19scfv基因通过连接肽克隆至fn7或fn10的n端和/或c端。获得的基因片段采用in-frame连接克隆至pfuse-higg1-fc2载体(invivogen,ca)的ecori和nhei酶切位点之间,测序验证。每个融合蛋白的核酸和氨基酸序列见seqidno:12--seqidno:62。

2.融合蛋白的表达和纯化

将实施例1构建好的融合蛋白表达载体瞬时转染freestylehek293细胞(gibco):将28mlfreestylehek293(3×107细胞/ml)接种至125ml细胞培养瓶,质粒用1mlopti-mem(gibco)稀释后加至1ml含60μl293fectintmtransfectionreagent(gibco)的opti-mem中,室温静置30min,将质粒-293fectinmixture加至细胞培养液中125rpm,37℃,5%co2培养。分别于转染后48h和96h收集细胞培养上清,proteina/g(thermofisherscientific,il)或ni-nta(genscript)纯化后,sds-page检测。检测结果见图5和图6.

3.体外活性研究

3.1促nfs-60细胞增殖实验

对实施例2制备好的融合蛋白fn10-1g-gcsf、fn10-1a-gcsf、fn7-1a-gcsf、fn7-1g-gcsf、fn7-1g-gcsf-fc进行促进nfs-60细胞增殖活性的研究。

具体步骤包括:培养nfs-60(美国atcc)细胞(rpmi-1640培养基:含10%胎牛血清,0.05mm2-巯基乙醇,62ng/mlhmcsf(r&dsystem)。增殖活性实验开始前,rpmi-1640不完全培养基洗nfs-60细胞3次,rpmi-1640完全培养基(含10%胎牛血清,0.05mm2-巯基乙醇)调整细胞密度1.5×105cells/ml,并加至96孔板中(100ul/孔)。往96孔板加入梯度稀释的humangcsf(r&dsystem)、fn10-1g-gcsf、fn10-1a-gcsf、fn7-1a-gcsf、fn7-1g-gcsf、fn7-1g-gcsf-fc蛋白于37℃,5%co2培养。72h后加入1/10体积的alamarblue(invitrogen)37℃孵育4h,测定595nm荧光值。

结果见图7a-7c。融合蛋白fn7-1g-gcsf、fn7-1a-gcsf、fn7-1g-gcsf-fc促进nfs-60细胞的增殖能力明显强于人gcsf(见图7a),但是fn7-1g-gcsf和fn7-1a-gcsf之间、fn10-1g-gcsf和fn10-1a-gcsf之间促增殖能力没有明显差异,提示a-helix和gslinker对融合蛋白促增殖能力影响很小。

3.2促nb2-11细胞增殖实验

培养大鼠nb2-11细胞(sigma)(rpmi培养基:含10%马血清(lifetechnologies,ca),55μm2-巯基乙醇(lifetechnologies,ca)),调整细胞密度2.5×105cells/ml,铺于96孔板(200ul/孔)。往96孔板加入梯度稀释的hgh(r&dsystem)或fn7-1g-hgh-fc蛋白于37℃,5%co2培养。72h后每孔加20μlcellviabilityreagent(lifetechnologies),读取590nm荧光值(激发波长550nm)。

结果如图8所示。fn7-1g-hgh-fc表现出于类似hgh促nb2-11细胞增殖能力。

4.体外细胞毒性检测

4.1ldh释放实验

收集健康志愿者外周血,通过ficoll-hypaque(gehealthcare)梯度离心分离外周血单核细胞(pbmcs),将分离的pmbcs用rpmi1640/10%fbs完全培养基重悬。将pbmcs与固相结合的anti-cd3抗体(cloneokt3,ebiosciences)、2μg/mlanti-cd28抗体(clonecd28.2,ebiosciences)37℃孵育,48h后加入20u/mlil2(r&dsystems)刺激活化的t细胞扩增。

dmem完全培养基(含10%fbs,1%青/链霉素)培养mda-mb-468(atcc)、mda-mb-231(atcc)、mda-mb-435/her2(atcc)、nalm-6(atcc)和ht-29(atcc)细胞,胰酶消化后,与上述活化的t细胞以10:1(t细胞密度106个细胞/ml,靶细胞密度105个细胞/ml)的比例孵育,加入梯度稀释的scfv2-fn7-scfv1、fab2-lc-fn7-scfv1或scfv3-fn7-scfv1,37℃,5%co2培养24h,cytotox-96nonradioactivecytotoxicityassaykit(promega)检测培养上清中ldh含量。spectramax250读取490nm处od值。细胞毒性(%表示)计算如下:

%细胞毒性=(实验吸光度–自发杀伤吸光度平均值)/(最大杀伤吸光度平均值–自发杀伤吸光度平均值)

其中,“最大杀伤”为仅有靶细胞的上清中ldh含量;“自发杀伤”为含有靶细胞和效应细胞(t细胞)、不加融合蛋白的细胞上清中的ldh含量。

结果见图9和图10。scfv2-fn7-scfv1和fab2-lc-fn7-scfv1对her2阳性的靶细胞mda-mb-231,mda-mb-435/her2、sk-br-3细胞具有较强的杀伤作用,而对于her2阴性的细胞则无影响。scfv3-fn7-scfv1对cd19阳性的nalm-6细胞具有较强的杀伤作用,对cd19阴性的ht-29细胞则几无影响。

4.2荧光染色

收集健康志愿者外周血,ficoll-hypaque(gehealthcare)梯度离心分离外周血单核细胞(pbmcs),rpmi1640/10%fbs完全培养基重悬。将pbmcs与固相结合的anti-cd3(cloneokt3,ebiosciences)、2μg/mlanti-cd28(clonecd28.2,ebiosciences)37℃孵育,48h后加入20u/mlil2(r&dsystems)刺激活化的t细胞扩增。

dmem完全培养基(含10%fbs,1%青/链霉素)培养nalm-6细胞,将nalm-6细胞铺于24孔板,与上述活化的t细胞以5:1(t细胞密度5*105cells/ml,靶细胞密度105cells/ml)的比例孵育,加入梯度稀释的scfv3-fn10-scfv1、scfv3-fn7-scfv1或bite,37℃,5%co2培养24h。用celltracerorangecmradye(lifetechnolgoy)染色后,在fitc(forcsfe)滤光片下观察荧光,统计活的nalm-6b细胞数。

结果如11所示。scfv3-fn7-scfv1和scfv3-fn10-scfv1均能有效抑制nalm-6细胞的活性,其效果与bite一致。

6.小鼠体内药代动力学

6.1fn7-1g-gcsf-fc

cd1小鼠(每组3只)静脉(i.v.)或皮下注射(s.c.)fn7-1g-gcsf-fc(2mg/kg)。注射后0-14天每日取血,利用anti-humaniggfc抗体(kpl)和anti-hgcsfantibody(abbiotec)抗体行elisa检测。以注射30min后的血样elisa读数为第一个时间点。

结果见图12。fn7-1g-gcsf-fc无论是皮下注射(s.c.)还是静脉注射(i.v.),其在体内的浓度缓慢下降,提示其具有较长的半衰期。

6.2anti-her2/anti-cd3或anti-cd19/anti-cd3

0.2mganti-her2/anti-cd3(scfv2-fn7-scfv1,fab2-lc-fn7-scfv1)或0.2mganti-cd19/anti-cd3bsabs(scfv3-fn7-scfv1)(溶于pbs,ph7.4)静脉注射cd1小鼠(每组6只)。分别于5min、15min、30min、1h、2h、4h、6h、8h、24h、32、48h后眼窝采集尾肝素抗凝血75ul,干冰保存。实验室将抗凝血12000rpm离心3min,取血浆铺于96孔板中,固相结合的herbb2-fc(r&dsystems)一抗和hrp抗人kappa(abcam)二抗检测血浆中融合蛋白的浓度。取最后4个时间点数据放入一级方程a=a0e-kt(a0为初始浓度,t为时间,k为一级速率常数)计算消除半衰期。

结果如图13a和图13b所示,scfv2-fn7-scfv1、fab2-lc-fn7-scfv1和scfv3-fn7-scfv1在血浆中的浓度随时间推移下降,其半衰期在20-30h之间,远远高于、比blinatumomab(bite)的半衰期(1.5-2.1h)。

7.小鼠体内药效动力学

7.1fn7-1g-gcsf-fc

单一剂量人gcsf(10ug/kg)或fn7-1g-gcsf-fc(50ug/kg)皮下注射balb/c小鼠(每组3只),注射后0-21天采集血样,fitcanti-cd45(miltenyibiotec)、peanti-cd11b(miltenyibiotec)和apcanti-ly-6g(bdbiosciences)抗体流式检测中性粒细胞的比例。

结果如图14所示。gcsf在注射后1天内血液中中性粒细胞急剧上升,之后迅速下降,第6天即检测不到中性粒细胞;而fn7-1g-gcsf-fc注射组血液中性粒细胞的变化趋势与gcsf一致,但是注射后6天至21天,中性粒细胞比例与第6天基本持平,提示fn7-1g-gcsf-fc持续发挥作用。

7.2anti-her2/anti-cd3

融合蛋白抑制荷瘤小鼠肿瘤块能力的检测在6-8周雌性nod-scid-γ小鼠(nod.cg-prkdcscidil2rgtm1wjl/szj;jacksonlaboratory)和人乳腺癌细胞her22+(mda-mb-453)、her21+(mda-mb-435)]上进行。

her22+肿瘤模型:5×106mda-mb-453细胞重悬于50%matrigel(bdbioscience)后皮下注射小鼠右胁腹,注射后第二天,腹腔注射2×107新鲜制备的pbmcs;同时,用固相结合的anti-cd3抗体(cloneokt3,ebioscience)、2μg/mlanti-cd28抗体(clonecd28.2,ebioscience)、50iu/ml重组人il-2(r&dsystems)体外刺激pbmcs。肿瘤细胞接种后第9天和第12天,腹腔注射2×107体外活化的t细胞;当肿瘤块大小达到200-300mm3时,每天静脉注射scfv2-fn7-scfv1、fab2-lc-fn7-scfv1(1mg/kg)或生理盐水,连续10天。每天测量小鼠体重。

her21+肿瘤模型:5×106mda-mb-435细胞重悬于50%matrigel(bdbioscience)后皮下注射小鼠右胁腹,注射后第二天,腹腔注射2×107新鲜制备的pbmcs。肿瘤细胞接种后第9天和第12天,腹腔注射2×107体外活化的t细胞;当肿瘤块大小达到200-300mm3时,每天静脉注射scfv2-fn7-scfv1、fab2-lc-fn7-scfv1(1mg/kg)或生理盐水,连续10天。每天测量小鼠体重卡钳测量所有实验小鼠肿瘤块大小,每周二次。按下式计算肿瘤体积:肿瘤体积=宽度*长度*高度

结果如图15所示。scfv2-fn7-scfv1和fab2-lc-fn7-scfv1处理后,mda-mb-453(her22+)荷瘤小鼠和mda-mb-435(her21+)荷瘤小鼠的肿瘤块明显缩小,且其体重没有明显变化,提示这些融合蛋白不仅具有较好的抑瘤活性而且具有较好的安全性。

7.3anti-cd19/anti-cd3bsabs

anti-cd19/anti-cd3bsabs抑制荷瘤小鼠肿瘤块的能力检测在nsg小鼠和nalm-6细胞上进行。

nsg小鼠接种5x105nalm-6细胞(表达gfp)(day0)。接种6天后,静脉输注4x107pbmc,6h后继续静脉输注scfv3-fn7-scfv1。每天测量小鼠体重,当小鼠体重比实验前减少超过15%时,处死小鼠。ivis测量肿瘤负荷,并以目标区域(roi,regionofinterest)的半径表示。

结果如图16所示,输注scfv3-fn7-scfv1后,荷瘤nsg小鼠肿瘤半径明显缩小,而pbs输注组其肿瘤持续增大,提示scfv3-fn7-scfv1抑瘤效果显著。

8.大鼠体内药代动力学

将fn7-1g-gh-fc静脉(i.v.)或者皮下注射(s.c.)sd雌性大鼠(每组3只)。尾静脉或隐静脉采集肝素抗凝血,采血时间如下:30min、1h、2h、4h、6h、24h、48h、3d、4d、6d、8d、10d、12d和14d。离心后取血浆,-80℃保存备用。血浆hgh含量采用hghhumandirectelisakit(lifetechnology)检测,具体步骤如下:用山羊抗人iggfc(abcam,ma)包被maxisorbelisa板(nunc),37℃孵育1h,5%bsa封闭,加入梯度稀释的血浆室温孵育1h。洗去未结合的血浆,加入一抗生物素标记的抗hgh多抗(r&dsystems,mn)孵育1h,洗板3次;加入链亲和素-hrp缀合物(thermofisherscientific,il)室温孵育1h,洗板三次;加quantabla荧光elisa底物(thermofisherscientific,il),spectramax检测荧光信号。根据标准曲线(横坐标为hgh浓度、纵坐标为荧光信号值)计算血浆中hgh的含量。药代动力学参数采用modelingprogramwinnonlin(pharsight)估算。

结果如图17所示。fn7-1g-gh-fc无论是皮下注射(s.c.)还是静脉注射(i.v.),其在体内停留的时间比较长,分别达到10天和14天,提示其具有较长的半衰期。

9.大鼠体内药效动力学

将humangh(0.1mg/ml,每天给药)及不同浓度fn7-1g-hgh-fc(0.5mg/kg、2.5mg/kg、5.0mg/kg;每周二次)皮下注射垂体切除的雄性sd大鼠(共8只)皮下注射给药,每天测体重。

结果如图18所示,pbs注射的sd大鼠其体重几乎变化很小,而不同浓度fn7-1g-hgh-fc每周二次注射的小鼠体重变化与0.1mg/kghgh每天给药的小鼠体重变化趋势一致,提示fn7-1g-hgh-fc可达到hgh类似的效果。

10.热力学稳定性测试

根据厂家提供的说明,利用荧光为基础的蛋白热量转移(fluorescence-basedproteinthermalshiftassay)(appliedbiosystems)测试fn10、fn10-1a-gcsf、fn10-1g-gcsf、scfv2-fn7-scfv1、scfv3-fn7-scfv1、scfv3-fn10-scfv1及fab2-lc-fn7-scfv1的热力学稳定性。将样品(0.5mg/ml,溶于pbs)与ptsdye(溶于ptsbuffer)混合,appliedbiosystemsviia7real-timepcr仪检测tm值。
