本发明涉及分子生物技术领域,特别涉及一种观赏性鱼类黑金红头鼠线粒体全基因组序列的扩增引物和扩增方法。
背景技术:
黑金红头鼠,学名内格罗河兵鲶(corydorasduplicareus),隶属辐鳍鱼纲鲶形目美鲶科兵鲶属。原产地是巴西境内nobuaoba北方8公里处的rioporanga,是一种优良的观赏性鱼类。本种延续红头鼠群特有的淡雅体色,以及令人一眼即可看出特色差异的隐眼线与背部线条而极具观赏价值。成熟雌性拥有橄榄状的体型,不但腹部线条向左右两侧扩张,同时从侧面观察,更可见到雌鱼随生殖巢逐渐发育,而拥有足以撑起头部与尾端的便便大腹,最大体长在50-55mm之间。其喜好酸性软水,生性较敏感,所以饲养底层宜放置细软的河沙为主,同时在水流与光照控制上,需妥善搭配具有隐蔽性的造景素材,提供一个稳定安静的环境。目前国内引进大量鲶科鱼类作为观赏品种,但大多命名比较混乱,对物种的鉴定不准确。此外,现阶段对黑金红头鼠的研究非常少,特别是在分子生物学方面没有确切数据及分析。因此,对黑金红头鼠线粒体全基因组序列进行扩增和分析,可为鼠鱼的品种鉴定和资源保护提供科学依据。
技术实现要素:
本发明所要解决技术问题是:针对上述现有技术的不足,提供一种观赏性鱼类黑金红头鼠线粒体全基因组序列的扩增引物和扩增方法,通过22对特异性引物对黑金红头鼠基因组进行pcr扩增,获得黑金红头鼠线粒体全基因组序列,为观赏鱼类品种鉴定、资源保护提供了分子生物学信息。
为解决述技术问题,本发明采用的技术方案是:一种黑金红头鼠线粒体基因组全序列的扩增引物,该扩增引物包括22对引物对:
第1对引物对的上游引物f1为:5’–caggcatcaggcacaatcat–3’,下游引物r1为:5’–ccggtacacttaccatgttacgac–3’;
第2对引物对的上游引物f2为:5’–gcacgaagtgggaagaaatg–3’,下游引物r2为:5’–ggcccactgaagcggtaatc–3’;
第3对引物对的上游引物f3为:5’–gctggttgcctaagaagtgg–3’,下游引物r3为:5’–tggtcgccccaaccgaagat–3’;
第4对引物对的上游引物f4为:5’–tgatctgcccgtgcagaagc–3’,下游引物r4为:5’–tgggtgtggcgagaaatagg–3’;

上述扩增引物是根据genbank数据库中收录的黑躯兵鲶(corydorasrabauti)线粒体全基因组序列设计得到。
本发明同时提供一种对黑金红头鼠线粒体基因组全序列扩增的方法,该方法是以提取的黑金红头鼠肌肉组织总dna为模板,分别用上述的22对引物对进行pcr扩增,扩增产物经1.0%琼脂糖凝胶电泳检测后进行测序及序列拼接。
上述提及的pcr的反应体系为50μl,其中:1μldna模板、1μl10μm上游引物、1μl10μm下游引物、1μl10mmdntp、5μl10×不含mg2+的pcrbuffer、5μl25mmmgcl2、0.5μl5u/μltaq酶及35.5μl无核酸酶水。pcr扩增条件为:95℃预变性3min,然后94℃变性45s,56℃退火1min,72℃延伸90s,共35个循环,最后72℃延伸8min。
本发明还提供根据上述扩增方法获得的黑金红头鼠线粒体基因组全序列,该线粒体基因组长度为16667bp,全基因组序列包含2个核糖体rna基因,13个不含内含子的蛋白编码基因,22个转运rna基因和2个非编码区(控制区和轻链复制起始区)。
具体实施方式
为了更好地解释本发明,以下结合具体的实施例进一步阐明本发明的主要内容,但本发明的内容不仅仅局限于以下实施例。
实施例1
1.材料与方法
1.1实验材料获取
从湖南省长沙市扬帆观赏鱼市场购买黑金红头鼠,按dna提取样品处理要求采集鱼背部肌肉,-20℃保存备用。
1.2肌肉基因组dna提取
按上海生工生物工程有限公司dzup基因组dna抽提试剂操作手册要求提取黑金红头鼠线粒体基因组dna,具体操作步骤如下:
a)取25-50mg的黑金红头鼠肌肉组织加液氮碾磨成粉末,加入到1.5ml的离心管中。加入1mldzup,震荡混匀,室温放置5-10min至完全裂解。震荡的时候应充分,要使溶液中无明显的组织团块即可。
b)将上一步获得裂解组织的离心管放入离心机以10000rpm/min室温离心10min,然后将上清液转移到新的1.5ml的离心管中。此步骤主要用来除去不溶的组织碎片和水解的rna等杂质。
c)加入500ul的无水乙醇,颠倒混匀5-8次,室温放置2-3min,如有沉淀产生,用移液枪头将沉淀的dna挑取到新的1.5ml离心管中。如无法挑取,8000rpm室温离心5min,弃上清液。
d)加入1ml75%的乙醇,颠倒1-3min,以12000rpm/min离心2min,弃上清液,多次重复,从而使沉淀悬浮起来。
e)开盖室温倒置5-10min至残留的乙醇挥发。
f)得到的dna用无菌蒸馏水进行溶解,-20℃保存备用。
1.3引物设计与pcr扩增
1.3.1引物设计
引物设计根据genbank收录的黑躯兵鲶线粒体基因组全序列(corydorasrabauti,收录号:ab054128.1)信息设计特异性引物,特异性引物由上海铂尚生物技术公司合成,共22对,具体如下:
第1对引物对的上游引物f1为:5’–caggcatcaggcacaatcat–3’(如seqidno.1所示),下游引物r1为:5’–ccggtacacttaccatgttacgac–3’(如seqidno.2所示);
第2对引物对的上游引物f2为:5’–gcacgaagtgggaagaaatg–3’(如seqidno.3所示),下游引物r2为:5’–ggcccactgaagcggtaatc–3’(如seqidno.4所示);
第3对引物对的上游引物f3为:5’–gctggttgcctaagaagtgg–3’(如seqidno.5所示),下游引物r3为:5’–tggtcgccccaaccgaagat–3’(如seqidno.6所示);
第4对引物对的上游引物f4为:5’–tgatctgcccgtgcagaagc–3’(如seqidno.7所示),下游引物r4为:5’–tgggtgtggcgagaaatagg–3’(如seqidno.8所示);

1.3.2pcr扩增
a)pcr反应体系(总体积50μl):1μldna模板、1μl上游引物(10μm)、1μl下游引物(10μm)、1μldntp10mm、5μl10×pcrbuffer(不含mg2+)、5μlmgcl2(25mm)、0.5μltaq酶(5u/μl)及35.5μl无核酸酶水(nuclease-freewater)。
b)pcr反应程序:95℃预变性3min,然后94℃变性45s,56℃退火1min,72℃延伸90s,共35个循环,最后72℃延伸8min。
c)pcr产物电泳检测:pcr扩增产物经1%琼脂糖凝胶电泳分离,90v电压电泳50min,sybrgreen染色后,凝胶成像系统进行条带分析并拍照保存。
1.4序列测定与拼接
1.4.1序列测定
将上一步电泳检测条带大小符合预期、带型单一的产物送上海铂尚生物技术公司测序,测序引物为片段扩增引物,双向测通。
1.4.2序列拼接与分析
测序所得各片段直接运用geneiousr11软件进行拼接和基因注释,ncbi在线多序列比对。
2.结果
经geneiousr11软件分析可知,黑金红头鼠线粒体全基因组序列长度为16667bp(如seqidno.45所示)。全基因组序列包含2个核糖体rna基因,13个不含内含子的蛋白编码基因,22个转运rna基因和2个非编码区(控制区和轻链复制起始区),与其他大多数鱼类的研究结果相同。
ncbi比对分析黑金红头鼠和黑躯兵鲶线粒体基因组,二者相似性为92%,存在1174个突变位点。
以上所述的实施例只是本发明的一种较佳的方案,并非对本发明做任何形式上的限制,在不超出权利要求所记载的技术方案的前提下,还有其他的变体和改型。
