本发明涉及:引起在植物中产生2n配子的TDM基因中的显性突变;包含所述突变的植物;以及它们在植物育种中的应用。本发明还涉及这样的植物,在该植物中,TDM基因中的显性突变与以下基因的失活组合:参与植物中减数分裂重组的基因,和参与减数分裂期间着丝粒的单极取向的基因。这些产生未减数分裂配子的植物也可用于植物育种。
背景技术:
减数分裂是有性繁殖的真核生物(诸如,大多数的开花植物)生命周期中的关键步骤。
在正常的减数分裂中,染色体首先复制,产生成对的姐妹染色单体。在该轮复制之后,进行两轮分裂,称为减数第一次分裂和减数第二次分裂。在减数第一次分裂期间,同源染色体重组并分离在两个细胞隔室(compartiment)中,这个两个细胞隔室各自都包含染色体的一个完整的单倍体内容物。在减数第二次分裂中,由减数第一次分裂产生的两组染色体进一步分裂,使姐妹染色单体分离开。因此,由该分裂产生的四个孢子是单倍体(n),并且携带重组的遗传信息。
相比之下,在二倍体细胞的有丝分裂期间,染色体进行复制,姐妹染色单体分离产生子代细胞,该子代细胞是二倍体(2n)的并且在遗传学上与原始细胞相同。
已经示出,由减数分裂期间的异常现象得到的异常配子可用于几种感兴趣植物(包括作物)的遗传改良(供参考,参见例如,Ramanna&Jacobsen,Euphytica 2003,133,3-18)。具体地,2n配子和未减数分裂配子(apomeiotic gamete)可用于产生多倍体植物,或用于使不同倍性水平的植物,例如四倍体作物与其二倍体野生近缘种杂交,以便在植物育种方法中使用它们的遗传多样性。它们也可用于遗传作图的方法。未减数分裂配子对于产生无融合生殖植物(即能够从胚珠的母体组织形成种子的植物)也是感兴趣的,从而产生作为母本的遗传克隆的子代。
2n配子(也称为二倍体配子)是具有体细胞染色体数,而不是配子体染色体数的配子。引起2n配子形成的异常情况特别包括异常胞质分裂,第一次减数分裂或第二次减数分裂的遗漏(skip),或纺锤体异形(供参考,参见V Veilleux,Plant Breeding Reviews,1985,3,252-288,or Bretagnolle&Thompson,New Phytologist,1995,129,1-22)。这些异常情况导致不同类别的未减数的配子。例如,第一次减数分裂的遗漏产生第一次分裂恢复(FDR)的配子,而第二次减数分裂的缺少产生第二次分裂恢复(SDR)的配子。已经报道了在各种植物物种中能够产生2n配子的许多突变体。然而,还未对这些植物中参与二倍体配子形成的突变进行表征。
未减数分裂配子是遗传学上与原始细胞相同的配子,保留亲本的所有遗传信息。未减数分裂配子的产生是无融合生殖的关键因素之一(Bicknell&Koltunow,Plant Cell,2004,16,S228-45)。虽然,超过400种植物是无融合生殖的,但这些植物中包括很少的作物物种。此外,通过杂交来引入这一性状的尝试已失败了(“The Flowering of Apomixis:From Mechanisms to Genetic Engineering”,2001;Editor:Savidan et al.;Publisher:CIMMYT,IRD,European Commission DG VI(FAIR),MEXICO,2001.Spillane et al.,Sexual Plant Reproduction,2001,14,179-187)。
迄今为止,已经证实了只有少数基因与2n或未减数分裂花粉的形成有关。
AtPS1(拟南芥平行纺锤体,Arabidopsis thaliana parallel spindles)的失活产生二倍雄孢子,从而产生具有重组遗传信息的可育的二倍花粉粒和在子代中产生自发三倍体植物(WO 2010/004431;d’Erfurth et al.,PLoS Genet.,2008,4,e1000274)。
TAM(缓慢异步的减数分裂,也称为CYCA1;2)或OSD1(第二次分裂的消失)的失活导致在减数第一次分裂之后过早地退出减数分裂,从而产生具有重组遗传信息的二倍体孢子和SDR配子(d’Erfuth et al.,PLoS Genet.,2010,6,e1000989;d’Erfuth et al.,PLoS Biol.,2009,7,e1000124;WO 2010/079432)。
SPO11-1编码植物中进行有效减数分裂重组所必需的蛋白,并且它的抑制消除重组和配对(Grelon et al.,Embo J.,2001,20,589-600);而Rec8(At2g47980)编码着丝粒在减数分裂期间的单极取向所必需的蛋白(Chelysheva et al.,J.Cell.Sci.,2005,118,4621-32),并且它的抑制改变染色单体的分离。Atspo11-1突变体经历不平衡的第一次分裂,随后进行第二次分裂,产生不平衡的孢子并且是不育的。Atspo11-1/Atrec8双突变体经历有丝分裂样分裂(而不经历正常的第一次减数分裂),随后进行不平衡的第二次分裂,产生不平衡的孢子并且是不育的(Chelysheva et al.,J.Cell.Sci.,2005,118,4621-32)。
在osd1/spo11-1/rec8三重突变体中,spo11-1和rec8突变的存在导致有丝分裂样的第一次减数分裂,并且osd1突变的存在阻止发生第二次减数分裂。因此减数分裂完全替换成有丝分裂,且不会影响随后的有性生殖过程。因此,将osd1/spo11-1/rec8突变体命名为MiMe(有丝分裂而不是减数分裂)(d’Erfuth et al.,PLoS Biol.,2009,7,e1000124和WO 2010/079432)。由MiMe突变体获得的孢子和配子在遗传上与原始细胞相同。
迄今为止,能够产生2n配子或未减数分裂配子的植物的工程因此限于有限数目的基因。
因此,为了增加可以经修饰以产生高频的2n配子或未减数分裂配子的基因的数目,需要参与植物中的这些配子形成的其它基因。
TDM(三次分裂突变,THREE-DIVISION MUTANT)基因,也被命名为TDM1、MS5(蛋白雄性不育,PROTEIN MALE STERILE 5)或POLLENLESS 3编码属于植物中保守的小蛋白家族的蛋白。拟南芥(Arabidopsis thaliana)的TDM基因的序列能够在TAIR数据库中以登录号At4g20900来获得,或在GenBank数据库中以登录号NC_003075.7来获得。它编码434个氨基酸(aa)的蛋白,其序列在所附的序列表中表示为SEQ ID NO:1。
TDM基因被描述为在减数分裂结束时退出减数第二次分裂所需的基因。TDM突变导致多分体(polyad)的形成,以及由于在正常的减数第一次分裂和减数第二次分裂后进入异常的第三次减数分裂而引起的雄性不育。据表明,tdm突变体携带突变,该突变产生编码在其C末端缺少305或112个氨基酸的截短的TDM蛋白的基因(Bulankova et al.,The Plant Cell,2010,22,3791-3803;Cromer et al.,PLoS Genet.,2012,8,e10028652012;Glover et al.,The Plant Journal,1998,15,345-356;Ross et al.,Chrom.Res.,1997,5,551-559;WO/9730581)。
与此相比,如本文所示,本发明人已经确定了TDM基因中的显性突变,该显性突变使得在减数第二次分裂之前提前退出减数分裂,从而产生具有重组遗传信息的二倍体雄SDR配子和二倍体雌SDR配子以及二倍体孢子。此外,他们已经表明,在spo11-1 rec8双突变体的TDM基因中引入显性突变产生了MiMe突变体。
本发明人因此鉴定出了在植物中参与2n配子和未减数分裂配子形成的另一个基因。
与参与二倍体配子产生的其它突变相比(这些突变是隐性的,因此需要初生突变体(突变杂合)的额外自花授粉步骤来获得突变纯合的植物),TDM中的显性的突变不需要这一步骤。携带TDM基因中的显性突变的初生突变体能够产生2n配子。

因此,本发明提供了一种用于获得产生第二次分裂恢复(SDR)2n配子的植物的方法,
其中,所述方法包括提供基因内包括显性突变的植物,所述基因在此指TDM基因,编码在此被指定为TDM蛋白的蛋白;
其中,当所述植物为芸苔属Brassica spp时,所述蛋白与SEQ ID NO:1的TDM蛋白的第1至286位氨基酸残基具有至少75%序列一致性;或者,当所述植物不同于芸苔属Brassica spp时,所述蛋白与所述残基具有至少30%序列一致性,并且所述蛋白的前60个氨基酸包括在此被指定为TPP/Q模体的模体X1X2X3,其中,X1为苏氨酸(T),X2为脯氨酸(P),以及X3为脯氨酸(P)或谷氨酰胺(Q);以及
其中,所述显性突变使所述植物具有产生SDR 2n配子的能力。
在下面的描述中,使用标准的单字母氨基酸代码。
TDM基因和蛋白质序列能够在公共数据库中获得,诸如但不限于,Plaza数据库(http://bioinformatics.psb.ugent.be/plaza/)和phytozome网络门户http://www.phytozome.net/(phytosome v9.1)。
在使用具有默认参数的T-Coffee(v6.85)(http://toolkit.tuebingen.mpg.de/t_coffee)进行序列比对之后,对SEQ ID NO:1的第1至286位残基计算TDM蛋白的蛋白序列一致性;使用Bioedit 7.2.5由该比对获得百分比一致性(Hall,T.A.,1999.BioEdit:a user-friendly biological sequence alignment editor and analysis program for Windows 95/98/NT.Nucl.Acids.Symp.Ser.41:95-98)。
每个物种具有一个或两个TDM基因,通常具有一个(图1和图2,以及表I)。取决于物种,TDM蛋白由约400个氨基酸至约850个氨基酸组成(图1)。根据图1中所示出各种被子植物物种的TDM蛋白的比对表明,TDM蛋白的前半部分是保守的。特别地,所有TDM蛋白的前60个氨基酸都包括保守的TPP/Q模体(图1)。
在使用具有默认参数的T-Coffee(v6.85)(http://toolkit.tuebingen.mpg.de/t_coffee)进行多重序列比对之后,计算各种被子物种的TDM蛋白与SEQ ID NO:1的TDM蛋白的第1至286位残基的百分比序列一致性。使用Bioedit 7.2.5由该比对获得一致性矩阵(Hall,TA,1999.BioEdit:a user-friendly biological sequence alignment editor and analysis program for Windows 95/98/NT.Nucl.Acids.Symp.Ser.41:95-98)。结果示于下表I中。

与SEQ ID NO:1的TDM蛋白的第1至286位氨基酸残基具有至少30%序列一致性的序列与SEQ ID NO:1的TDM蛋白的第1至286位氨基酸残基具有至少50%的序列相似性。因此,除了芸苔属Brassica spp.植物以外的其它植物的TDM蛋白可替代地被限定为与SEQ ID NO:1的TDM蛋白的第1至286位氨基酸残基具有至少50%的序列相似性,并且包括在该蛋白的前60个氨基酸中的TPP/Q模体。
根据本发明产生的SDR 2n配子可用于2n配子的所有常见应用中,例如用于生产多倍体植物,或允许不同倍性水平的植物之间进行杂交。它们也可用于遗传作图的方法,例如WO 2006/094774中所公开的“反向子代作图”的方法中。
根据用于获得产生第二次分裂恢复2n配子的植物的方法的优选实施方式,当所述植物不同于芸苔属Brassica spp时,所述蛋白与SEQ ID NO:1的TDM蛋白具有至少35%的序列一致性,并且逐渐优选地,具有至少40%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%或98%的序列一致性;或者,当所述植物为芸苔属Brassica spp时,所述蛋白与SEQ ID NO:1的TDM蛋白具有至少80%的序列一致性,并且逐渐优选地,具有至少85%、90%、95%或98%的序列一致性。根据用于获得产生第二次分裂恢复2n配子的植物的方法的另一优选实施方式,所述TPP/Q模体位于所述蛋白的SEQ ID NO:1的第16~18位的区域中。
根据本发明,所述显性突变在TDM基因或TDM转基因的等位基因内,其中,所述TDM基因或转基因来自任何植物物种,例如表I中提及的那些物种。因此,能够产生SDR 2n配子的植物能够通过TDM基因的定向诱变或随机诱变来获得,或者通过遗传转化来获得。
根据本发明的用于获得产生第二次分裂恢复2n配子的植物的方法的优选实施方式,所述方法包括:
提供在TDM基因的等位基因内具有所述显性突变的植物,所述植物对于该突变是杂合的或纯合的。
TDM基因的诱变可以是定向的或随机的。在随机诱变(例如通过EMS诱变)之后,筛选期望基因内的突变体。用于高通量诱变和筛选的方法是本领域可知晓的。作为实例,可以提及TILLING(定向诱导基因组局部突变(Targeting Induced Local Lesions IN Genomes),由McCallum et al.,Plant Physiology,2000,123,439-442所述)。利用标准技术进行定向诱变,该标准技术是本领域所已知的并且利用同源重组,优选与核酸酶例如TALEN或CRISPR组合。
根据本发明的用于获得产生第二次分裂恢复2n配子的植物的方法的另一优选实施方式,所述植物是转基因植物,并且所述方法包括:
a)用包含DNA构建体的载体转化至少一个植物细胞,所述DNA构建体包括具有所述显性突变的TDM基因;
b)培养所述转化的植物细胞,以使在其基因组中具有转基因的植物再生,该转基因包括所述DNA构建体。
DNA构建体包含TDM基因,该TDM基因可来自与其将引入的植物相同的物种,或来自不同的物种。
在TDM基因内的突变中,可以基于对于该突变是杂合的植物的表型性状,来确定导致产生SDR 2n配子的能力的那些突变:这些植物可以形成至少5%,优选至少10%,更优选至少20%,还更优选至少50%,并且高达100%的双孢体,作为减数分裂的产物。
可替代地,可通过它们恢复拟南芥(A.thaliana)spo11/rec8双突变体的可育力的能力,来确定TDM基因内,导致突变体或转基因植物具有产生SDR 2n配子的能力的显性突变,其中可育的spo11/rec8/tdm三重突变体对于TDM突变是杂合的,如本申请的实施例中所证实的,拟南芥(A.thaliana)spo11/rec8双突变体用于筛选显性突变,随后将该显性突变引入到感兴趣的植物中。
根据本发明的用于获得产生第二次分裂恢复2n配子的植物的方法的另一优选实施方式,所述显性突变包括保守TPP/Q模体的至少一个残基的突变或由保守TPP/Q模体的至少一个残基的突变组成。
上述突变可以是置换、插入或缺失,优选置换或缺失。
在更优选的实施方式中,所述显性突变包括T残基的突变和/或它相邻的P残基的突变,或由T残基的突变和/或它相邻的P残基的突变组成。TP是潜在的磷酸化位点。本申请的实施例证实了T16或P17残基的突变能够显著地提前退出减数分裂。不希望受理论的束缚,本发明人认为鉴于这些结果,TDM通过磷酸化来调节,以确保减数第一次分裂至减数第二次分裂的过渡。
因此,突变有利地是消除所述模体的T残基磷酸化的突变,即,破坏TP磷酸化位点的突变。
所述突变有利地是所述T残基和/或P残基被不同的残基置换,例如T被A置换,P被L置换。可替代地,所述突变是缺失T残基和P残基,并且最终缺失所述T残基和/或P残基两侧的额外残基,例如缺失1个至10个,优选1个至5个,甚至更优选1个或2个残基。
如上所限定,本发明的另一方面涉及包括TDM基因的DNA构建体,所述TDM基因具有使突变体/转基因植物具有产生SDR 2n配子的能力的显性突变。TDM基因可来自与其将引入的植物相同的物种,或来自不同的物种。DNA构建体包括可表达形式的TDM基因。优选地,TDM基因位于启动子的转录控制下,该启动子在植物细胞中有功能。启动子可以是TDM基因启动子,诸如所述TDM基因的内源启动子,或在植物中有功能的另一启动子。
在本领域中,具有大量供选择的适于在植物中表达异源基因的启动子。
它们可以例如获自植物、植物病毒或细菌(诸如农杆菌(Agrobacterium))。它们包括:组成型启动子,即在大多数组织和细胞中以及在大多数环境条件下具有活性的启动子;以及,组织特异性启动子或细胞特异性启动子,仅仅或主要在特定组织或特定细胞类型中具有活性;和,诱导型启动子,该诱导型启动子通过物理刺激或化学刺激,诸如由于线虫感染而产生的物理刺激或化学刺激而被激活。对启动子进行选择以便在性母细胞中具有功能。
常用于植物细胞中的组成型启动子的非限制性实例是花椰菜花叶病毒(CaMV)35S启动子、Nos启动子、1,5-二磷酸核酮糖羧化酶/加氧酶(rubisco)启动子、木薯叶脉花叶病毒(CsVMV)启动子。
可用于本发明的器官或组织特异性启动子特别包括能够赋予减数分裂相关表达的启动子,诸如DMC1启动子(Klimyuk&Jones,Plant J,1997,11,1-14)。
本发明的DNA构建体通常还包括转录终止子(例如,35S转录终止子、胭脂碱合酶(Nos)转录终止子或TDM基因终止子)。
本发明还包括重组载体,所述重组载体包含本发明的DNA构建体。通常,所述重组载体还包括一个或多个标记基因,允许对转化的宿主进行选择。
合适载体的选择,以及用于将DNA构建体插入到其中的方法是本领域普通技术人员所熟知的。载体的选择取决于预期的宿主以及所述宿主的预期的转化方法。在本领域中,对于许多植物物种,双子叶植物或单子叶植物,有多种用于植物细胞或植物的遗传转化方法。作为非限制性实例,可提及的是:病毒介导的转化,通过显微注射、电穿孔进行的转化,微粒介导的转化,农杆菌(Agrobacterium)介导的转化等。
本发明还提供了包括本发明的重组DNA构建体的宿主细胞。所述宿主细胞可以是:原核细胞,例如农杆菌(Agrobacterium)细胞;或者真核细胞,例如通过本发明的DNA构建体遗传转化的植物细胞。构建体可被瞬时表达;它也可被并入到稳定的染色体外复制子中,或整合到染色体中。
本发明人还发现,通过使TDM突变中的显性突变与两个基因的失活相组合,来得到产生未减数分裂配子的MiMe突变体,该两个基因中的一个基因对于减数分裂重组的起始是必需的并且选自spo11-1、SPO11-2、PRD1、PRD2(AT5G57880)、PRD3/PAIR1和DFO(AT1G07060),另一个基因为REC8。
由MiMe突变体产生的未减数分裂配子可以与SDR 2n配子相同的方式,用于生产多倍体植物或用于不同倍性水平的植物的杂交。它们对于生产无融合生殖植物也是感兴趣的,即能够从胚珠的母体组织形成种子的植物,从而产生为母本的遗传克隆的子代。
因此,本发明的另一目的是用于获得产生未减数分裂配子的植物的方法,其中所述方法包括:
a)提供植物,所述植物包括如上定义的在TDM基因中的显性突变;
b)在所述植物中,抑制参与植物中起始减数分裂重组的第一蛋白,所述蛋白选自以下蛋白:

使用在默认参数下的BLASTP程序,计算spo11-1、spo11-2、prd1、PRD2、DFO、PAIR1或REC8蛋白的在此所提供的蛋白序列一致性和相似性的值。使用评分矩阵BLOSUM62,进行相似性计算。
可通过消除、阻断或降低其功能,或者有利地通过阻止或下调相应基因的表达,来获得对上述SPO11-1、SPO11-2、PRD1、PRD2、PAIR1、DFO或Rec8蛋白的抑制。在申请WO2010/00431中详细公开了SPO11-1、SPO11-2、PRD1、PRD2、PAIR1、DFO或Rec8蛋白以及所述蛋白的抑制。
例如,所述蛋白的抑制可以通过诱变相应的基因或其启动子,并选择所述蛋白的活性部分或完全丧失的突变体来获得。所述抑制公开在WO 2010/00431的第5页的最后一段开始至第6页的第4段结束,以及第6页的最后一段,这些内容通过引用并入本文中。

f)使步骤e)的植物自花授粉,以获得对于TDM基因内的突变是纯合的,对于选自SPO11-1、SPO11-2、PRD1、PRD2、DFO或PAIR1基因的基因内的突变是纯合的,以及对于REC8基因内的突变的是纯合的植物。
根据本发明的用于获得能够产生未减数分裂配子的植物的方法的另一优选实施方式,所述植物是转基因植物,并且所述方法包括:
a)用含有本发明的包含如上定义的TDM基因中的显性突变的DNA构建体的载体,含有靶向选自SPO11-1、SPO11-2、PRD1、PRD2、DFO或PAIR1的基因的DNA构建体的载体,以及含有靶向REC8基因的DNA构建体的载体转化至少一个植物细胞;
b)培养所述转化的植物细胞,以使在它的基因组中具有含所述DNA构建体的转基因的植物再生。
在TDM基因中包括显性突变的DNA构建体的表达,为所述转基因植物提供了产生2n SDR配子的能力。包括TDM基因中的显性突变的DNA构建体基因,靶向选自SPO11-1、SPO11-2、PRD1、PRD2、DFO或PAIR1的基因的DNA构建体,以及靶向REC8基因的DNA构建体的共表达,使由这三种基因编码的蛋白下调,并且为所述转基因植物提供产生未减数分裂配子的能力。

实验步骤
1.生长条件和基因分型
在21℃下,在16小时至18小时光周期和70%相对湿度下,如先前所述在温室中(Vignard et al.,PLoS Genet.,2007,3,1894-1906)或者如前所述离体在拟南芥Arabidopsis培养基上(Estelle and Somerville,Mol.Genet.,1987,206,200-206)上培养Arabidopsis拟南芥植物。
如先前所述(d’Erfurth et al.,PLoS Biol.,2009,7,e1000124)对spo11-1-3rec8-2植物进行基因分型。如Cromer et al.,PLoS Genet.,2012,8,e1002865所述对tdm-3植物进行基因分型。
2.EMS诱变和突变鉴定
如先前所述(Crismani et al.,Science,2012,336,1588-1590)进行EMS诱变。通过HigSeqTM2000(Illumina)进行全基因组测序。与拟南芥Arabidopsis thaliana TAIR10(哥伦比亚栽培品种)的对照基因组相比,生成SNP列表。
3.细胞学和倍性分析
使用由d'Erfurth等人(PLoS Genet.,2008,4,e1000274)描述的技术,对雄性减数分裂产物进行观察、染色体分散和倍性测量。
4.定向诱变构建体和植物转化
使用TDM U(5'-GACATCGGCACTTGCTTAGAG-3';SEQ ID NO:36)和TDM L(5'-GCGATATAGCTCCCACTGGTT-3';SEQ ID NO:37)通过PCR扩增TDM基因组片段。扩增覆盖ATG前的986个核苷酸和终止密码子后的537个核苷酸。通过GatewayTM技术(Invitrogen),将PCR产物克隆到pDONR207TM载体(Invitrogen)中,以产生pENTR-TDM;使用Stratagene QuickChangeTM定点诱变试剂盒,根据制造商的说明对pENTR-TDM进行定向诱变。用于产生TDM的突变形式的诱变引物为SEQ ID NO:38至SEQ ID NO:41:

以及它们各自的反向互补引物。为了产生用于植物转化的双元载体,使用Gateway TM系统的二元载体pGWB1进行LR重组反应(Nakagawa et al.,Journal of Bioscience and Bioengineering.2007,104,34-41)。对野生型植物以及分离成spo11-1或rec8或tdm-3突变的植物群,使用农杆菌介导的花浸渍法(Clough,S.J.and Bent A.F.,Plant J.,1998,16,735-743)转化得到的二元载体pTDM,pTDM-P17L,pT16A和pTDM-Y14A。在含有20mg/L潮霉素的琼脂板上对转化的植物进行选择。
实施例1:TDM中的显性突变导致提早退出减数分裂
为了鉴定控制减数分裂进程的新基因,基于以下观点来设计遗传筛选:导致遗漏第二次减数分裂的突变,诸如osd1和cyca1;2/tam将恢复仅在第二次减数分裂中具有不平衡的染色体分离缺陷的突变体的可育力(d’Erfurth et al.,PLoS Biol.,2009,7,e1000124;d’Erfurth et al.,PLoS Genet.,2010,6,e1000989和WO 2010/079432)。这是spo11rec8双突变体的情况,其中第一次减数分裂类似于有丝分裂(姐妹染色单体平衡分离至相对极),但是第二次分裂是不平衡的且产生非整倍体的配子,因此具有非常有限的可育力(图5)(Chelysheva et al.,J.Cell.Sci.,2005,118,4621-32)。在OSD1(d’Erfurth et al.,PLoS Biol.,2009,7,e1000124 2009)或CYCA1;2/TAM(d’Erfurth et al.,PLoS Genet.,2010,6,e1000989)中的导致在减数第二次分裂前退出减数分裂的突变,确实能够恢复spo11-1 rec8的可育力。因此,基于spo11-1 rec8的可育力的恢复来运行遗传筛选,旨在鉴定出赋予比osd1或tam还相似缺陷的突变体。尽管它们具有减数分裂分离的缺陷,spo11-1 rec8植物产生了经甲基磺酸乙酯(EMS)诱变的足够的剩余种子。假定对EMS突变是杂合的M1植物进行自花授粉,并且在5个批次中收获以产生M2家族。筛选了与spo11-1 rec8非诱变对照相比,可育力增加的约2000个M2家族(400个批次)。
三个批次分离具有增加可育力的植物。基因分型证实,它们是spo11-1 rec8突变体,这表明了其是真正的抑制因子。通过甲苯胺蓝染色对雄性减数分裂产物进行的分析示出,在所有三种情况下,可育植物产生孢子的二分体(如在osd1或cyca1;2/tam中所观察到的),而不是四分体(如在野生型中所观察到的),表明了在这些植物中并没有发生第二次减数分裂(图3)。候选基因(CYCA1;2/TAM和OSD1)的测序确定了CYCA1;2/TAM在三个家族中的两个中的隐性突变。所确定的突变是外显子7(TAIR10chr1:29082522C>T)中的剪接位点,和5'UTR区域中的引入上游框外起始密码子(TAIR10chr1:29084174G>A)的突变。互补试验表明它们是等位基因,证实了CYCA1;2中的突变引起二分体表型和可育力的恢复。第三个家族(spo11rec8(s)-40)在OSD1和CYCA1;2中没有突变,是本研究的重点。
出乎意料地,染色体分散表明四种植物是四倍体的(图3)。这表明因果突变(causal mutation)是显性的,并且导致在M1植物的雄性和雌性器官中均产生二倍体配子。与野生型相比,以~100X覆盖度对两种姐妹植物的大部分所进行全基因组测序显示出1144个SNP的存在。然而,仅15个SNP呈现为纯合子。这些少数纯合子SNP分散在基因组中,表明它们在诱变前就存在于spo11-1rec8株系中,而不是由于EMS诱导的突变所固定的。几乎所有检测到的突变都是杂合子这一事实进一步表明因果突变是显性的。该突变会在M1植物中显型表达,在与spo11-1 rec8突变组合下,导致二倍体克隆配子的产生(如在spo11-1rec8osd1三重突变体中观察到的)(MiMe,d’Erfurth et al.,PLoS Biol.,2009,7,e1000124和WO 2010/079432),因此四倍体M2植物中维持了来自M1植物的EMS诱导的突变的杂合性。
然后,在杂合子SNP中寻找候选的因果突变。在这1129个突变中,预测341个突变将影响编码序列(无义、错义或剪接位点)。其中,TDM中导致氨基酸变化的突变(TDM-P17L)由于潜在的因果显性突变而成为良好候选者。之前已证实,TDM对于减数第二次分裂结束时退出减数分裂是必需的。即使在tdm敲除突变体中观察到的减数分裂缺陷(额外的一轮分裂)与(spo11rec8(s)-40)缺陷显著不同,TDM中的显性突变仍然是(spo11rec8(s)-40)中的因果突变的潜在候选者。
为了验证该假设,产生含有TDM基因(包括启动子和终止子)的基因组克隆,并使其突变以重新产生在筛选中所鉴定的突变(TDM-P17L),其中TDM基因能够补充tdm-3突变体(n=8个转化体,100%四分体)。当引入spo11-1rec8植物中时,TDM-P17L克隆恢复了初代转化体的可育力(n=2/3,spo11-1 rec8:0.1个种子/果实(n=197),spo11-1 rec8 TDM-P17L#15:25个种子/果实(n=15),spo11-1 rec8 TDM-P17L#67:48个种子/果实(n=10)),并产生二分体(图4,表II)。这表明TDM中的突变确实是spo11rec8(s)-40中的因果显性突变。对spo11-1rec8TDM-P17L转化体中减数分裂染色体分散的分析显示出,有丝分裂样的第一次分裂,其中10个单价体在中期I时对齐,并且姊妹染色单体在后期I分离,且不进行第二次分裂(图5)。接下来,研究了spo11-1rec8 TDM-P17L后代的倍性水平。在自交子代中,仅发现四倍体(4n)(表III)。当spo11-1rec8 TDM-P17L花粉用于使野生型植物受精时,所有得到的子代都是三倍体(表III)。当spo11-1 rec8 TDM-P17L胚珠经野生型花粉粒受精时,仅发现三倍体植物(表III)。这证实了被TDM-P17L转化的spo11-1 rec8突变体产生高水平的雄性和雌性(100%)有丝分裂样来源的孢子,其导致功能性二倍体配子。
当引入到野生型植物和tdm-3突变体中时,TDM-P17L基因组克隆通过产生二分体来修饰两种基因型的减数分裂表型(图4,表II)。

产生二分体的TDM-P17L植物显示出野生型第一次分裂并且不发生减数第二次分裂(图5),从而形成2n配子,以及来自osd1和cyca1;2/tam之一的表型重现(d’Erfurth et al.,PLoS Biol.,2009,7,e1000124和WO 2010/079432;d’Erfurth et al.,PLoS Genet.,2010,6,e1000989)。在TDM-P17L植物的后代中测量倍性水平(表III)。在自交子代中,发现了四倍体和三倍体。当TDM-P17L胚珠经野生型花粉粒受精时,分离出二倍体植物和三倍体植物(表III)。

总之,tdm-p17L显性突变赋予了与隐性osd1或tam突变类似的减数分裂缺陷,使在第二次分裂之前提前退出减数分裂,从而产生二倍体的雄性和雌性配子。
TDM属于植物中保守的小家族蛋白。例如,拟南芥Arabidopsis基因组含有与TDM显示出显著序列相似性的5个其他基因(图2)。这些类TDM基因的功能是未知的。蛋白序列的分析表明,因果突变在仅TDM蛋白亚家族中保守的小结构域中,所述TDM蛋白亚家族通常每个植物物种含有一个或两个基因(图1)。Pro17氨基酸以及相邻的Thr16氨基酸是绝对保守的(图1)。这就限定了T16上的最小共有磷酸化位点。为了对此进行验证,通过用不可磷酸化的氨基酸(TDM-T16A)来置换可磷酸化的氨基酸,并通过删除整个保守结构域(TDM-Δ14_19)来建立基因组TDM基因的两个其它潜在的磷酸化丢失形式。当引入到野生型植物中时,TDM-T16A和TDM-Δ14_19都以显性方式产生了二分体表型,重现了TDM-P17L的作用(表II;图4)。此外,当引入到tdm-3突变体时,TDM-Δ14_19也显示二分体表型(表II)。然而,当引入到野生型时,TDM酪氨酸14(TDM-Y14A)(结构域的保守性稍低的氨基酸)的突变不能产生二分体表型,但能够补充tdm-3突变(表II)。总之,TDM-P17L、TDM-T16A和TDM-Δ14-19突变的表达同样能够显性地提早退出减数分裂。由于TP是潜在的磷酸化位点,这个结果表明TDM可能受到磷酸化的调节以确保减数第一次分裂至减数第二次分裂的过渡。
