本发明属于基因工程领域,公开了大豆与本氏烟草中四个植物nac转录因子nac089基因及其表达载体在植物抵抗疫霉侵染中的应用。
背景技术
辣椒疫霉菌是一种广谱的病原菌,能够在辣椒、烟草等多种经济作物上造成严重危害。辣椒疫霉隶属于卵菌,该类病原菌虽然在形态上与真菌相似,但在进化上却和硅藻及蓝藻关系较近,为茸鞭生物界,因此一般真菌杀菌剂往往对大豆疫霉和其它卵菌无效。植物抗病品种的应用,由于克服了药害以及环境污染等问题,成为农业生产上实现抗病的重要手段。
植物与病原菌的长期协同进化过程中,植物为了抵抗病原菌,进化出了大量的抗病相关基因,其中包括许多在抗病过程中起作用的nac转录因子。nac089是其中的一个比较特殊的转录因子,其不但含有nac结构域,还含有跨膜结构域,在内质网介导的免疫系统中起重要作用。将nac089导入到作物中,可以提高作物对辣椒疫霉的抗性。以本氏烟草为例,在本氏烟草中瞬时表达大豆和本氏烟草中的nac089基因,可以显著提高本氏烟草对辣椒疫霉的抗性。在没有特别好的防治辣椒疫霉的方法的现状下,对于nac089的克隆对于利用基因工程手段培育具有辣椒疫霉抗性的作物品种具有重要意义。
技术实现要素:
本发明的目的为了弥补目前防治作物疫病的不足,提供了大豆与本氏烟草中四个植物nac转录因子nac089基因及其表达载体在植物抵抗疫霉侵染中的应用。
本发明的目的可以通过以下技术方案实现:
来自本氏烟草(nicotianabenthamiana)和大豆(glycinemax)williams82两种植物材料的四个nac转录因子nac089基因,nbnac089-1、nbnac089-2,gmnac089-1,gmnac089-2,所述nbnac089-1、nbnac089-2,gmnac089-1和gmnac089-2基因的核苷酸序列依次如seqidno.1—4所示。
上述四个nac转录因子nac089基因编码的蛋白质为nbnac089-1、nbnac089-2,gmnac089-1,gmnac089-2,其氨基酸序列为seqidno.5—8。
含有上述nac转录因子nac089基因的表达盒、重组表达载体、转基因细胞系或转基因重组菌。
含有上述nac转录因子nac089基因的重组表达载体为pbingfp2:nbnac089-1,pbingfp2:nbnac089-2,pbingfp2:gmnac089-1和pbingfp2:gmnac089-2。构建所述nac转录因子基因nbnac089-1、nbnac089-2,gmnac089-1,gmnac089-2的重组表达载体pbingfp2:nbnac089-1,pbingfp2:nbnac089-2,pbingfp2:gmnac089-1,pbingfp2:gmnac089-2。以植物表达载体pbingfp2为出发载体,将nbnac089-1、nbnac089-2,gmnac089-1,gmnac089-2基因分别插入pbingfp2的sam1酶切位点获得。
所述nac转录因子nac089基因及其表达载体可以用于构建抗疫霉(如辣椒疫霉和大豆疫霉等)作物品种。
一种构建抗疫霉作物品种的方法,将上述的基因或重组表达载体导入作物植株中,经过抗性筛选得到阳性转化植株,获得抗疫霉作物品种。
本发明优点和积极效果表现在:
本发明从大豆与本氏烟草中克隆到了四个植物nac转录因子nac089基因,将其插入表达载体pbingfp2。将所述载体导入作物中,可以提高作物对疫霉的抗性。所述nac转录因子可以用于基因工程,构建抗疫病的作物,同时为我们在农业生产中制定抗病策略、控制病害提供有力指导。

具体实施方式
下面结合实施例对本发明做进一步的说明,但不限制本发明的保护范围。
实施例1在本氏烟中瞬时表达nbnac089-1、nbnac089-2,gmnac089-1,gmnac089-2提高本氏烟对辣椒疫霉的抗性
1.供试菌株
供试菌株辣椒疫霉lt263由南京农业大学植病系植物与疫霉互作实验室保存(本领域技术人员广泛使用的常规菌株)。菌株保存于10%v8固体培养基,保存温度为10℃。
2.供试本氏烟草幼苗的准备
本氏烟草(nicotianabenthamiana)播种于装有蛭石(2-4mm)的塑料花盆(d=10cm)里,放到14h光照(强光照射)/10h黑暗的温室中生长。使其生长7天,生长出两片真叶后,将其移栽至蛭石黑土体积比例为5:1的培养土里生长30天,取相同3,4,5叶位的叶片用作表达gfp和nac089蛋白,然后接种辣椒疫霉。
3.供试大豆幼苗的准备
鉴别寄主大豆播种于装有蛭石(2-4mm)的塑料花盆(d=10cm)里,每盆种15粒种子,放到14h光照(强光照射)/10h黑暗的温室中生长,7天后两片真叶长出后留10颗苗用于接种。所用品种为williams。
4.提取大豆和本氏烟草rna,并获得nac089基因的cdna
收集大豆和本氏烟草的叶片,采摘后液氮速冻后放入研钵冷却研磨。参照天根总rna抽提纯化试剂盒推荐方法进行大豆和本氏烟草总rna提取和纯化。参照诺维赞公司rna反转录试剂盒提供的方法,以oligo(dt)为引物反转录合成cdna。以上述cdna为模板,用相应上下游引物进行pcr扩增,引物序列依次如seqidno.9—16所示。
pcr反应体系为:2.5μl10×pcr反应缓冲液;1.5μl1.5mmmgcl2;0.5μl2.5mmdntps;0.25μltaqdna聚合酶(5.0u/μl);0.5μl引物;0.5μl模板;无菌水补足至25μl。
反应程序为:94℃5min预变性;94℃变性15s,58℃退火15s,72℃延伸1:30min,35个循环;72℃延伸10min;4℃保存。
电泳:用1%琼脂糖凝胶电泳检测pcr产物,利用紫外凝胶成像仪上切取目的片段(大小约为1023bp)。pcr产物通过takara凝胶回收试剂盒回收,并送金斯瑞公司进行测序。扩增获得四个nac转录因子nac089基因,nbnac089-1、nbnac089-2,gmnac089-1,gmnac089-2,测序结果显示所述nbnac089-1基因的序列如seqidno.1所示、所述nbnac089-2基因的序列如seqidno.2所示,所述gmnac089-1基因的序列如seqidno.3所示,所述gmnac089-2基因的核苷酸序列如seqidno.4所示。四个nac转录因子nac089基因nbnac089-1、nbnac089-2,gmnac089-1和gmnac089-2编码的蛋白质的氨基酸序列依次如seqidno.5—8所示。
5、构建pbingfp2:nbnac089-1,pbingfp2:nbnac089-2,pbingfp2:gmnac089-1,pbingfp2:gmnac089-2表达载体
将pbingfp2空载体质粒sami进行酶切。利用诺韦赞公司的同源重组酶将cdna和载体连接。
反应体系如下:2μl10×ceⅱ反应缓冲液;2μl上述pcr回收产物;2μl上述酶切后的空载体;1μl同源重组酶;无菌水补足至10μl。
反应程序为:37℃条件下反应30min。将连接产物转化大肠杆菌dh5α,在kan抗性培养基上筛选转化子,挑取阳性克隆提取质粒进行菌落pcr鉴定。将鉴定到的阳性克隆测序,正确即获得了pbingfp2:nbnac089-1,pbingfp2:nbnac089-2,pbingfp2:gmnac089-1,pbingfp2:gmnac089-2表达载体。
6、获得农杆菌感受态细胞
在lb固体培养基平板上划线培养农杆菌gv3101菌株,28℃条件下倒置培养18-20h;将单菌落置于lb液体培养基(含有100mg/lrif),28℃条件下振荡培养18-20h;100倍体积的不含抗生素的lb液体培养基(含有100mg/lrif)加入菌液中,28℃条件下振荡培养od600至0.3-0.5;冰上冷却后,4℃条件下4000r/min离心5min,弃上清;加入20mmol/l的cacl2溶液使沉淀悬浮,4℃条件下4000r/min离心5min,弃上清;加入20mmol/l的cacl2溶液再次使菌体沉淀悬浮备用。
7、pbingfp2:nbnac089-1,pbingfp2:nbnac089-2,pbingfp2:gmnac089-1,pbingfp2:gmnac089-2表达载体和pbingfp2空载体转化农杆菌
利用电转法将pbingfp2:nbnac089-1,pbingfp2:nbnac089-2,pbingfp2:gmnac089-1,pbingfp2:gmnac089-2表达载体和pbingfp2空载体转化农杆菌细胞;利用70%酒精冲洗电击杯三次,然后完全风干酒精。将100ng表达载体加入农杆菌感受态,冰上放置30min。将农杆菌感受态细胞转移至电击杯中,利用2.5kv的电压进行电击转化。电击完成后将农杆菌感受态细胞加入到400μllb液体培养基中,28℃振荡培养2h。吸取20μl菌液,均匀涂抹在含有50mm卡那霉素和25mm的利福平固体lb培养基平板上,28℃倒置培养48h。挑取单菌落进行菌落pcr,获取阳性克隆。
8、在本氏烟草中瞬时表达gfp和nac089蛋白
将上述阳性克隆放置于lb液体培养基中,28℃震荡培养30h。将菌液收集,用8000rpm离心2min,利用10mmmgcl2清洗三次。最后用10mm的mgcl2稀释菌液至od600值0.3。将菌液与含有沉默抑制子p19的菌液1:1体积比混合。用1ml注射器注射分别注射满同一本氏烟草叶片的左面和右面。
9、瞬时表达gfp和nac089蛋白的本氏烟抗病性检测
将确认表达蛋白的叶片剪下,放置于滤纸保湿的托盘内。用7mm打孔器在辣椒疫霉lt263的平板生长边缘打孔,将打下的菌碟菌面向下得左右对称放置在本氏烟叶片的左右两侧。将托盘放置于25℃培养箱,黑暗,36h。取出叶片,然后用手持紫外荧光仪照射叶片。颜色变深的区域即为辣椒疫霉lt263的侵染区域。用量尺测量侵染区域的直径,表达gfp蛋白的叶面为对照,比较四种蛋白对本氏烟抗性的影响。发现来自大豆和本氏烟草的四种nac089蛋白显著提高了本氏烟草对辣椒疫霉lt263的抗性。
