一种生物合成牛心果碱的微生物及其方法与流程

本发明属于微生物代谢工程领域,具体涉及改造微生物代谢途径,从单糖或甘油直接合成牛心果碱的方法。



背景技术:

苄基异喹啉类生物碱包含2500多个种类,许多苄基异喹啉类生物碱,如:吗啡、小檗碱和诺斯卡品等在医学领域具有广阔的应用前景与经济地位。牛心果碱是苄基异喹啉类生物碱的共同中间体。

目前已商品化的苄基异喹啉类生物碱主要通过植物提取。由于植物生长周期长及苄基异喹啉类生物碱含量低,利用植物提取苄基异喹啉类生物碱成本较高。通过生物技术手段,实现微生物发酵单糖或甘油合成苄基异喹啉类生物碱共同中间体牛心果碱,可有效的降低成本,为规模化生物合成苄基异喹啉类生物碱提供了可行的道路。



技术实现要素:

本发明的目的在于提供一种微生物体内合成牛心果碱的方法,在微生物内构建一条由单糖或甘油转化到牛心果碱的生物合成途径。

为实现上述目的,本发明提供了在大肠杆菌合成牛心果碱的方法,生物合成路径如图1所示,主要步骤为:

(1)敲除基因构建菌株dy

以大肠杆菌mg1655菌株出发,依次敲除该菌株中七个醛还原酶基因(dkgb、yeae、yqhc、yqhd、dkga、yahk和yjgb),敲除上述7个基因的菌株命名为dy。

(2)构建产牛心果碱工程菌dy1

将来自大肠杆菌的解反馈抑制arogfbr(3-deoxy-d-arabinoheptulosonate-7-phosphatesynthetase,arog)基因和tyrafbr(chorismatemutase/prephenatedehydrogenase,tyra)基因及来自酿酒酵母脱羧酶kdc基因,重组到载体pbbr1mcs1上,得到载体pdy001。将来自大肠杆菌的hpabc(4-hydroxyphenylacetate3-hydroxylase,hpabc,由hpab和hpac组成)及经密码子优化人工合成的pseudomonasputida菌dodc(dopadecarboxylase,dodc)、papaversomniferum的cns((s)-norcoclaurinesynthase,cns)、papaversomniferum的6omt((rs)-norcoclaurine6-o-methyltransferase,6omt)、coptisjaponica的4omt(3'-hydroxy-n-methylcoclaurine4'-o-methyltransferase,4omt)和papaversomniferum的cnmt((s)-coclaurinen-methyltransferase,cnmt)基因,重组到载体pet28a(+)上,得到载体pdy002。将经密码子优化人工合成的mycobacteriummarinum菌car(carboxylicacidreductase,car)、bacillussubtilissubsp.subtilisstr.168菌的sfp(phosphopantetheinyltransferase,sfp)和大肠杆菌mg1655的aldh(aldehydedehydrogenase,aldh)基因重组到载体pet28b-psc101,得到载体pdy003。将载体pdy001、pdy002和pdy003转入到敲除七个醛还原酶基因的大肠杆菌dy中,实现上述12个基因在大肠杆菌中的高量表达,得到大肠杆菌工程菌株dy2。敲除七个醛还原酶基因用于阻断中间体4-羟基苯乙醛和3,4-二羟基苯乙醛被还原为副产物4-羟基苯乙醇和3,4-二羟基苯乙醇。表达解除反馈抑制的arogfbr基因和tyrafb基因用于加强大肠杆菌胞内对羟基苯丙酮酸和酪氨酸的合成。表达hpab、hpac、kdc、aldh和car用于催化对羟基苯丙酮酸转化为3,4二羟基苯乙醛。表达sfp用于活化car酶。表达hpab、hpac和dodc用于催化酪氨酸转化为3,4二羟基苯乙胺,表达cns、6omt、4omt和cnmt用于随后催化3,4二羟基苯乙醛与3,4二羟基苯乙胺两个产物缩合修饰生成牛心果碱。上述构建的dy1工程菌可有效的将单糖或甘油转化为牛心果碱。

具体而言,本发明一方面涉及一种微生物,其特征在于所述微生物不含有或者敲除dkgb、yeae、yqhc、yqhd、dkga、yahk和yjgb基因,高表达或者导入arogfbr、tyrafb、kdc、hpab、hpac、dodc、car、sfp、aldh、cns、4omt、6omt和cnmt基因。

进一步的,所述dkgb基因具有seqidno:1所示的核苷酸序列或者所述dkgb基因编码seqidno:2所示的氨基酸序列;

所述yeae基因具有seqidno:3所示的核苷酸序列或者所述yeae基因编码seqidno:4所示的氨基酸序列;

所述yqhc基因具有seqidno:5所示的核苷酸序列或者所述yqhc基因编码seqidno:6所示的氨基酸序列;

所述yqhd基因具有seqidno:7所示的核苷酸序列或者所述yqhd基因编码seqidno:8所示的氨基酸序列;

所述dkga基因具有seqidno:9所示的核苷酸序列或者所述dkga基因编码seqidno:10所示的氨基酸序列;

所述yahk基因具有seqidno:11所示的核苷酸序列或者所述yahk基因编码seqidno:12所示的氨基酸序列;

所述yjgb基因具有seqidno:13所示的核苷酸序列或者所述yjgb基因编码seqidno:14所示的氨基酸序列;

所述arogfbr基因具有seqidno:15所示的核苷酸序列或者所述arogfbr基因编码seqidno:16所示的氨基酸序列;

所述tyrafb基因具有seqidno:17所示的核苷酸序列或者所述tyrafb基因编码seqidno:18所示的氨基酸序列;

所述kdc基因具有seqidno:19所示的核苷酸序列或者所述kdc基因编码seqidno:20所示的氨基酸序列;

所述dodc基因具有seqidno:21所示的核苷酸序列或者所述dodc基因编码seqidno:22所示的氨基酸序列;

在本发明的一个优选实施方式中,微生物为大肠杆菌。

本发明另一方面还涉及上述微生物在生物合成牛心果碱中的应用。

作为微生物发酵发酵单糖或甘油合成牛心果碱的方法,所述的单糖为葡萄糖。

本发明的优点是在微生物中引入外源基因构建一条合成牛心果碱的代谢途径,最终实现以单糖或甘油为原料合成牛心果碱的目的,相比较与植物提取或化学合成可有效的降低成本。同时,微生物生长速度快,遗传操作技术成熟,且发酵过程不会对环境造成污染破坏。

具体实施方式

以下实施例用于进一步说明本发明,但不应理解为对本发明的限制。

实施例1构建产牛心果碱大肠杆菌工程菌

1.基因敲除菌株dy的构建

以pkd4质粒为模板,pcr扩增打靶片段,该片段包含(kanr序列和与待敲除目的基因两端互补的同源臂);将扩增的打靶片段电转到带有pkd46质粒的大肠杆菌中。利用pkd46质粒上带有的red同源重组系统,使待敲除目的基因与打靶片段进行重组,目的基因被抗性基因取代。随后将温敏质粒pcp20导入菌株,利用该质粒上的flp翻转酶去除目的菌株染色体上的抗性基因。利用上述方法依次敲除大肠杆菌mg1655菌株中的dkgb、yeae、yqhc、yqhd、dkga、yahk和yjgb基因,获得大肠杆菌dy。所述dkgb基因具有seqidno:1所示的核苷酸序列;所述dkgb基因具有与seqidno:2所示的氨基酸序列;所述yeae基因具有seqidno:3所示的核苷酸序列;所述yeae基因具有与seqidno:4所示的氨基酸序列;所述yqhc基因具有seqidno:5所示的核苷酸序列;所述yqhc基因具有与seqidno:6所示的氨基酸序列;所述yqhd基因具有seqidno:7所示的核苷酸序列;所述yqhd基因具有与seqidno:8所示的氨基酸序列;所述dkga基因具有seqidno:9所示的核苷酸序列;所述dkga基因具有与seqidno:10所示的氨基酸序列;所述yahk基因具有seqidno:11所示的核苷酸序列;所述yahk基因具有与seqidno:12所示的氨基酸序列;所述yjgb基因具有seqidno:13所示的核苷酸序列;所述yjgb基因具有与seqidno:14所示的氨基酸序列;敲除基因用的引物为:

2.载体pdy001的构建

解除反馈抑制的arogfbr和tyrafbr基因委托金唯智生物科技有限公司合成,以合成的基因为模板利用引物arogfbr-xbai(gtatctagaaagaggagatataatgaattatcagaacgacgatttacgc)和arogfbr-spei-bamhi(tatggatccactagtttacccgcgacgcgctttta)pcr扩增arogfbr基因,随后将扩增的片段以xbai和bamhi插入到pet28a(+)中得到载体pdy1。以合成的基因为模板,利用引物tyrafbrr-xbai(gtatctagaaagaggagatataatggttgctgaattgaccgcat)和tyrafbr-spei-sali(atatgtcgacactagtttactggcgattgtcattcgcc)pcr扩增来tyrafbr基因,随后将扩增的片段以xbai和sali双酶切,插入到以spei和sali双酶切的载体pdy1上,中得到载体pdy2。利用引物kdc-xbai(atctctagatttaagaaggagatataatggcacctgttacaattgaaaagttc)和kdc-nhei-bamhi(tctggatccgctagcctattttttatttcttttaagtgccgctg)pcr扩增来自酿酒酵母菌yph499的kdc基因,随后将扩增的片段以xbai和bamhi双酶切,将其插入以xbai和bamhi双酶切的载体pbbr1mcs1上,得到载体pdy3。利用xbai和sali双酶切pdy2得到arogfbr和tyrafbr表达框,将其插入以nhei和xhoi双酶切的载体pdy3,得到载体pdy001。所述arogfbr基因具有seqidno:15所示的核苷酸序列;所述arogfbr基因具有与seqidno:16所示的氨基酸序列;所述tyrafb基因具有seqidno:17所示的核苷酸序列编码;所述tyrafb基因具有seqidno:18所示的氨基酸序列;所述kdc基因具有seqidno:19所示的核苷酸序列编码;所述kdc基因具有seqidno:20所示的氨基酸序列。

3.载体pdy002的构建

dodc、cns、6omt、4omt和cnmt基因委托金唯智生物科技有限公司合成。以合成的基因为模板,利用引物dodc-xbai(tagtctagatttaagaaggagatataatgacccccgaacagttccg)/dodc-spei-bamhi(acaggatccactagtttagctgccgcctttaatcacgt)、cns-xbai(tactctagatttaagaaggagatataatgcgtaaagtgatcaaatatgatatgg)/cns-spei-bamhi(tcaggatccactagtttagctgcccagtaaacgaggaa)、6omt-xbai(ttgtctagatttaagaaggagatataatggagaccgttagcaaaattgacc)/6omt-spei-bamhi(gcaggatccactagtttagtacggatatgcttcaatcacgct)、4omt-xbai(tactctagatttaagaaggagatataatggccttccatggcaaggac)/4omt-spei-bamhi(tcaggatccactagtttaaggaaacacctcaatcacgctct)、cnmt-xbai(atctctagatttaagaaggagatataatgcagttaaaagaaaaagaggagctg)/cnmt-spei-bamhi(agaggatccactagtttattttttcttaaacagcagatgggttaac),分别pcr扩增dodc、cns、6omt、4omt和cnmt基因,随后将扩增的片段以xbai和bamhi插入到pet28a(+)中,依次得到载体pdy4、pdy5、pdy6、pdy7和pdy8。利用xbai和xhoi双酶切pdy5得到cns表达框,将其插入以spei和xhoi双酶切的载体pdy4上,得到载体pdy9。利用xbai和xhoi双酶切pdy6得到6omt表达框,将其插入以spei和xhoi双酶切的载体pdy9上,得到载体pdy10。利用xbai和xhoi双酶切pdy7得到4omt表达框,将其插入以spei和xhoi双酶切的载体pdy10上,得到载体pdy11。利用xbai和xhoi双酶切pdy8得到cnmt表达框,将其插入以spei和xhoi双酶切的载体pdy11上,得到载体pdy12。以大肠杆菌mg1655的基因组为模板,利用引物hpabc-xbai(tagtctagatttaagaaggagatataatgaaaccagaagatttccgcg)和hpabc-xhoi(aacgctcgagttaaatcgcagcttccatttcca),pcr扩增hpabc基因(包含hpab和hpac两个基因),随后将扩增的片段以xbai和xhoi双酶切,将其插入以spei和xhoi双酶切的载体pdy12上,得到载体pdy002。所述dodc基因具有seqidno:21所示的核苷酸序列;所述dodc基因具有与seqidno:22所示的氨基酸序列;所述cns基因具有seqidno:23所示的核苷酸序列;所述cns基因具有与seqidno:24所示的氨基酸序列;所述6omt基因具有seqidno:25所示的核苷酸序列;所述6omt基因具有与seqidno:26所示的氨基酸序列;所述4omt基因具有seqidno:27所示的核苷酸序列;所述4omt基因具有与seqidno:28所示的氨基酸序列;所述cnmt基因具有seqidno:29所示的核苷酸序列;所述cnmt基因具有与seqidno:30所示的氨基酸序列;所述hpab基因具有seqidno:31所示的核苷酸序列;所述hpab基因具有与seqidno:32所示的氨基酸序列;所述hpac基因具有seqidno:33所示的核苷酸序列;所述hpac基因具有与seqidno:34所示的氨基酸序列;

4.载体pdy003的构建

car基因委托金唯智生物科技有限公司合成。以合成的基因为模板,利用引物car-xbai(atctctagatttaagaaggagatataatgagtccgatcacacgcgag)/car-spei-bamhi(agaggatccactagtttacagcaggcctaacaggcg)pcr扩增car基因,随后将扩增的片段以xbai和bamhi双酶切,插入到以xbai和bamhi双酶切的pet28a(+)中,得到载体pdy13。以芽孢杆菌bacillussubtilissubsp.subtilisstr.168的基因组为模板,利用引物sfp-xbai(gtctctagatttaagaaggagatataatgaagatttacggaatttatatggaccg)/sfp-spei-bamhi(tgaggatccactagtttataaaagctcttcgtacgagaccattgt)pcr扩增sfp基因,随后将扩增的片段以xbai和bamhi双酶切,插入到以xbai和bamhi双酶切的pet28a(+)中,得到载体pdy14。以大肠杆菌mg1655的基因组为模板,利用引物aldh-xbai(atctctagatttaagaaggagatataatgaattttcatcatctggcttactg)/aldh-nhei-bamhi(tcaggatccgctagctcaggcctccaggctatcc)pcr扩增aldh基因,随后将扩增的片段以xbai和bamhi双酶切,插入到以xbai和bamhi双酶切的pet28a(+)中,得到载体pdy15。利用xbai和xhoi双酶切pdy14得到sfp表达框,将其插入以spei和xhoi双酶切的载体pdy13上,得到载体pdy16。利用xbai和xhoi双酶切pdy15得到aldh表达框,将其插入以spei和xhoi双酶切的载体pdy16上,得到载体pdy17。利用xbai和xhoi双酶切pdy17得到car、sfp和aldh表达框,将其插入以xbai和xhoi双酶切的载体pet28b-psc101上,得到载体pdy003。所述car基因具有seqidno:35所示的核苷酸序列;所述car基因具有与seqidno:36所示的氨基酸序列;所述sfp基因具有seqidno:37所示的核苷酸序列;所述sfp基因具有与seqidno:38所示的氨基酸序列;所述aldh基因具有seqidno:39所示的核苷酸序列;所述aldh基因具有与seqidno:40所示的氨基酸序列;

5.将重组载体pdy001和pdy002同时导入大肠杆菌dy中,得到dy1工程菌。该工程菌可实现arogfbr、tyrafbr、kdc、hpabc、dodc、cns、6omt、4omt和cnmt基因的高表达,可有效的将单糖或甘油转化为牛心果碱。

5.将重组载体pdy001、pdy002和pdy003同时导入大肠杆菌dy中,得到dy2工程菌。该工程菌可实现arogfbr、tyrafbr、kdc、hpabc、dodc、cns、6omt、4omt、cnmt、car、sfp和aldh基因的高表达。相较于dy1工程菌,进一步实现car、sfp和aldh基因高效表达的dy2工程菌,可提升的将单糖或甘油转化为牛心果碱的生物合成效率。

实施例2大肠杆菌工程菌dy1生产牛心果碱的发酵实验

1、将37度lb中过夜培养的大肠杆菌工程菌dy1,接种到50mlm9发酵培养基中(包含2%葡萄糖),进行30度摇瓶发酵,待菌体od600为0.8左右,加入0.1mmiptg进行目的基因的诱导表达。

2、发酵培养24小时后收集菌体,超声波破细胞,利用氯仿萃取产物。

3、利用旋转蒸发仪将溶剂氯仿旋干,再用一定量的氯仿溶解产物。

4、gc-ms(气相色谱-质谱联用仪)检测样品。

5、实验结果:大肠杆菌工程菌利用葡萄糖合成牛心果碱的gc-ms检测结果如图2a所示,牛心果碱gc-ms鉴定如图3所示,表明菌株dy1可有效利用葡萄糖或甘油合成牛心果碱。

实施例3大肠杆菌工程菌dy2生产牛心果碱的发酵实验

1、将37度lb中过夜培养的大肠杆菌工程菌dy2,接种到50mlm9发酵培养基中(包含2%葡萄糖),进行30度摇瓶发酵,待菌体od600为0.8左右,加入0.1mmiptg进行目的基因的诱导表达。

2、发酵培养24小时后收集菌体,超声波破细胞,利用氯仿萃取产物。

3、利用旋转蒸发仪将溶剂氯仿旋干,再用一定量的氯仿溶解产物。

4、gc-ms(气相色谱-质谱联用仪)检测样品。

5、实验结果:大肠杆菌工程菌dy2利用葡萄糖合成牛心果碱的gc-ms检测结果如图2b,表明菌株dy2可有效利用葡萄糖或甘油合成牛心果碱。

以上所述仅是本发明的优选实施方案,应当指出,对于本技术领域的技术人员,在不脱离本分明原理的前提下,还可以做出适当的改进,这些改进也应视为本发明的保护范围。
