SOX1突变蛋白、核酸分子、检测产品及应用的制作方法

本发明涉及生物检测领域,尤其涉及sox1突变蛋白、核酸分子、检测产品及应用。


背景技术:

1、鼻咽癌是好发于鼻咽侧壁和顶部的恶性肿瘤。鼻咽癌家族史一级亲属患病率比散发个体患病率高4-20倍。

2、癌症家族史是许多癌症公认的一种危险因素,有阳性癌症家族史的个体需要有效的遗传咨询和风险评估,但对鼻咽癌来说,目前还没有有效的筛查手段(如基因panel)用于该疾病的风险分层以及个体化预防。现有研究报道的鼻咽癌相关遗传易感基因如:hla某些基因型、tp53、tnfrsf19、3q26mecom、cdkn2a/2b以及5p15l 1l/tert会增加患鼻咽癌的风险,然而这些位点对鼻咽癌总体患病风险的贡献相对有限。

3、一些高外显率的胚系致病基因携带者发生遗传相关肿瘤的风险明显增加,如携带brca1/2胚系变异的女性发生乳腺癌和/或卵巢癌的风险高达80%以上。cdkn2a胚系变异携带者,发生遗传性黑色素瘤风险总体在58%~91%之间。apc胚系变异导致的家族性腺瘤性息肉病患者(familial adenomatous polyposis,fap),发展成结肠癌的几率为100%。rb胚系变异携带者发生遗传性视网膜母细胞瘤的风险高达90%,这类患儿通常具有发病早(1岁以内)和双侧发病特点。这些突变基因对肿瘤的遗传咨询以及风险评估具有重要作用。然而高外显率的鼻咽癌易感基因未见文献报道。


技术实现思路

1、通过全外显子测序、生物信息学分析以及家系共分离分析对典型鼻咽癌家系进行研究,显示该家族鼻咽癌符合显性遗传,初步确定发生该家族鼻咽癌的病因与sox1基因突变有密切关系。鉴于此,本发明提供了sox1突变蛋白、核酸分子、检测产品及应用,将对鼻咽癌的诊断与治疗提供一种全新的分子检测方法。
