雌激素受体雌激素受体  雌激素受体属类固醇类受体,纯化的受体相对分子质量为65000,第322~395位氨基酸为雌激素结合区,调控雌激素受体基因位于X性染色体,雌激素受体基因在第6对染色体,分A、B、C、D、E、F 6个区,C和E区分别为DNA和配基结合区。雌激素受体在羧基末端有配基结合区,其组成的氨基酸与雌激素、孕激素受体的配基结合区有许多相似性。其功能除与相应配基结合外,还具有二聚体化和核定位的功能。雌激素受体是一种酸性蛋白质,不溶于30%的硫酸铵溶液,对热极不稳定,未结合的受体比游离受体复合物更敏感。它不仅存在于细胞浆,也存在胞核内。其生物学特性为结合容量低、高亲和力、特异性强和组织特异性,如子宫、乳腺、阴道的性激素受体含量较非靶组织器官要高得多,目前已证实,骨骼、心脏、肌肉和大脑等组织均存在雌激素受体。在正常月经周期中,子宫内膜ER和PR呈周期性变化,增生期因E2的升调节作用逐渐增加,到增生晚期达高峰,分泌期因P的降调节作用ER和PR则下降。在病理情况下,ER含量会发生明显变化。
