6-巯基嘌呤分子式:C5H4N4S分子量:152.19CAS号:50-44-2性质:从水中析出者为微黄色棱状结晶一水合物(C5H4N4S·H2O,[6112-76-1]),在140℃失去结晶水。熔点313-314℃(分解)。溶于100份沸水,极微溶于冷水,几乎不溶于醇和醚。在碱笥溶液中缓慢分解,在空气及光照中颜色变深。无气味,味微苦。制备方法:由4,5-二氨基-6-羟基嘧啶(见1160)与甲酸环合6-羟基嘌呤(C5H4N4O),再与五硫化二磷经置换反应制得巯嘌呤。将4,5-二氨基-6-羟基嘧啶、甲酸加入反应锅中,搅拌加热溶解,升温回流4h,减压回收甲酸至尽。加入氢氧化钠溶液溶解,加活性炭脱色,过滤。滤液冷却并用冰醋酸调节pH至6,放置后过滤出结晶,即6-羟基嘌呤。将6-羟基嘌噙和五硫化二磷加入吡啶中搅拌溶解,在118℃反应4h,即生成巯嘌呤,经后处理精制而得成品,收率近60%。用途:该品为次黄嘌呤类干扰核合成的药物,又称抗代谢药物。阻止核酸的合成,制止癌细胞分裂繁殖,甚至因DNA合成受阻但蛋白质合成并不停止,因而发生不平衡生长,使癌细胞死亡。巯嘌呤属于周期特异性药物,主要作用于S期,对G1期也有延缓作用;对体液免疫和细胞免疫也有抑制作用。主要用于白血病。本品也是一种生化试剂。小鼠腹腔注射LD50为157mg/kg。
