SV40载体分子式:CAS号:性质:也可简称为。由于SV40病毒基因组具备:(1)由单一的共价闭合的环状DNA所组成;(2)含有多种功能的基因组的区域已正确定位;(3)病毒基因组能够自我复制。而可用作载体。野生型病毒DNA加入外源DNA后会导致DNA分子过大需缺失部分非必需DNA后亦用作载体。现在某些质粒型表达载体带有来自SV40的个别调控区,但不含病毒组的大部分编码区。通常采用的调控区是SV40DNA中的一个300bp的区段,位于病毒早期与晚期转录单位之间,含有一系列各不相同的顺式调控要素。这些调控要素包括:(1)DNA复制起始点;(2)早期与晚期mRNA转录的起始点和启动子;(3)能够激活复制起始点并可自动调节早期转录的T抗原结合位点;(4)可被细胞转录因子识别的一般由3个G/C丰富的21bp同向重复区组成的序列;(5)组成SV40早期增强子的两段72bp同向重复序列。除此之外,某些以SV40为基础的质粒载体因带有完整的SV40转录单位,激活源细胞SV40复制起始点,也可以编码SV40大T抗原。
