PAGE分子式:CAS号:性质:用聚丙烯酰胺为支持基质的电泳程序。一般说来,实现聚丙烯酰胺凝胶电泳有两种类型。(1)单向电泳:采用完整的蛋白质或用十二烷基磺酸钠(SDS)处理的蛋白质的单向泳动,在有凝胶的平板上平行分离(过去也有用在玻管中的圆筒形棒状凝胶进行电泳的,现已很少有人使用)。(2)双向电泳:首先用天然蛋白质进行分离,然后凝胶平板再用SDS处理,样品便在第二向得到分离。第一次分离了各种各样的蛋白质,因此第二向分离的是蛋白质亚基。主要优点是:(1)合成聚合物,故重复性良好;(2)分离能力好;(3)通过增减丙烯酰胺单体和交联剂(N,N′-亚甲基双丙烯酰胺)的浓度,可以调节凝胶的孔径大小;(4)操作简便、时间短;(5)化学性质稳定、机械性能好,柔软;(6)在酸性或碱性缓冲液中均可进行电泳,而且可加入两性电解质进行等电点电泳,可用含电解质表面活性剂(SDS)或非电解质表面活性剂(Np40、Tritonx-100等)的凝胶进行电泳,亦可使两者组合进行双向电泳等等,使用范围广泛,利用价值日益提高;(7)由于染色技术的进步,可以进行定量,也可检测出极微量的斑点(琼脂糖电泳)。
