生物素标记核酸探针技术分子式:CAS号:性质:这是一种非放射性标记技术。主要利用了生物素(biotin)和亲和素(avidin)这两种分子的独特性质。亲和素是一种碱性的四聚体糖蛋白,其分子量为68000,pI10.5。每个亚基都能结合一个生物素分子。生物素与亲和素的结合只需要生物素的脲基环部分。因此可将其戊酸侧链通过酰胺键与核酸分子相连,构成生物素标记的核酸探针、标记探针与固定在滤膜(硝酸纤维素滤膜或尼龙膜)上或固定在组织细胞或染色体原位的核酸分子的同源序列互补杂交。探针和待侧核酸在杂交前若为双链,应先变性成单链,然后用偶联有酶或荧光物质的亲和素的探针与待测核酸的片段互补杂杂,使杂交部位显色(加入相应的酶底物)或产生荧光。灵敏度可达1~10pg水平,与放射性探针法相近。如果进一步改进,甚至可超过后者。
