精子间接免疫荧光法精子间接免疫荧光法  诊法。免疫学检查方法之 一。用于不育症检查。①以志愿供者的精液02ml加入10~12ml生理盐水中离心,取其精子沉淀部分再置入05ml生理盐水内。②取一滴作玻璃涂片,并在空气内晾干30分钟,用纯甲醇固定30分钟,再晾干后置入pH为74的磷酸缓冲液内15分钟。③被测血清滴于上述制备的玻片上,室温下孵化30~60分钟后,用磷酸缓冲液漂洗3次,每次10分钟。④玻片上滴注 附有抗人体免疫球蛋白抗血清的异硫氰酸荧光素盐酸盐,室温下孵化30~60分钟,孵化后用磷酸缓冲液漂洗3次,每次10分钟。⑤荧光显微镜下观察结果。利用血清内的精子抗体,可使精子不同部位呈现不同免疫荧光素反应,通过与正常血清对比研究,以判断精子抗体的存在情况。
