基因分离分子式:CAS号:性质:从目的生物体中以DNA序列的形式分离出基因信息,其目的是要研究它的结构或在基因操作过程中把它插入一个载体。分离真核DNA通常是很困难的,甚至是不可能的。现常用反转录方法,即从mRNA开始,在反转录酶作用下,生成DNA,即cDNA,再将cDNA在DNA聚合酶作用下合成为双链DNA。这种cDNA不含内含子,但含有全部蛋白质(或多肽)的遗传信息。另一常用方法是在目的基因结构或蛋白质一级结构清楚的基础上,通过人工合成的引物用PCR法合成。再一方法是按既知序列,在核酸合成仪上合成,如序列太长可分成N段合成,然后体外退火,连接酶处理,从而合成一完整的基因DNA片段。对原核生物常用限制切取片段或PCR法制取。
