端粒酶分子式:CAS号:性质:一种以逆转录的方式合成真核细胞中线性染色体端粒DNA的特殊酶。该酶的辅基为一特定的RNA,端粒DNA的合成必须依赖酶分子中的辅基RNA为模板,通过逆转录而完成,其自身携带RNA作为产物的模板是区别于一般由纯蛋白组成的逆转录DNA聚合酶的主要特征。在逆转录时需要一个DNA引物,以便从5′→3′方向添加端粒的重复序列。底物是脱氧核苷三磷酸(dNTPs)。如在四膜虫(tetraphymena)中,四膜虫端粒酶催化反应式如下:染色体-GGGGTTGGGGTT(DNA引物)+4ndGTP(dNTPs)+2nTTP(dNTPs)→染色体-GGGGTTGGGGTT(GGGGTT)n(四膜虫端粒DNA)+6n焦磷酸端粒酶的活性在胚系细胞、癌化细胞中活跃;在体细胞,尤其经多次分裂后的体细胞,其活性降低。该酶的活性也可能是多细胞器官限制其细胞增殖的一种机制之一。
