核糖体失活蛋白分子式:CAS号:性质:又称核糖核酸N-糖苷酶,RNAN-糖苷酶。核糖体失活蛋白是一类专一性修饰真核或原核生物的核糖体高分子量RNA,从而成为抑制蛋白质生物合成的核糖体毒素,故可致细胞于死地。专一水解哺乳动物核糖体28S RNA中一个保守的腺苷酸的N-C糖苷键,释放一个腺嘌呤碱基使核糖体失活。除少数真菌和细胞外,迄今发现绝大多数存在于植物种子中。根据结构和功能的差异,核糖体失活蛋白目前被分为两类,即I型和Ⅱ型。I型由一条多肽链构成,分子量25 000~31 000。仅具失活多糖体的功能,它对离体细胞的蛋白合成比体内有较强抑制作用,属于碱性或强碱性蛋白质。属于I型核糖体失活蛋白的有天花粉蛋白(trichosanthin)等。Ⅱ型的核糖体失活蛋白由A和B两条肽链通过一对二硫键相连而成,分子量60 000~65 000,等电点6~8之间,A链作用与I型相近似,是RIP分子中的催化亚基,通过修饰真核糖体bos亚基抑制蛋白合成。B链相当于一个凝集素,可选择性地与细胞表面糖肽或糖脂受体结合,具有细胞识别功能,而且能协助A链通过膜进入细胞,定位于高尔基体上。蓖麻毒蛋白(ricin)、相思豆毒蛋白(abrin)是两种典型的Ⅱ型核糖体失活蛋白。核糖体失活蛋白在植物体内并不失活自身的核糖体,这说明它们在植物细胞体中不表现为RNA N-糖苷酶的活性。目前核糖体失活蛋白已成为研究核糖体拓扑结构和rRNA催化功能的一个很有效的分子探针之一;又鉴于它对细胞的剧毒性,人们用它们与单克隆抗体或某些激素或药物等结合构成“免疫毒素”,试图应用于治疗包括癌肿在内的多种疾病。
