静水式生物测试  测定污染物对水生生物的毒性的一种方法,即把受试生物置于不流动的试验溶液中,测定污染物浓度与受试生物中毒反应之间的关系,从而确定污染物的毒性。这种方法又称静水式试验。    影响测试的因素  在测试中,如其他试验条件不变,被测物浓度和试验溶液水质的变化会直接影响测定的结果。如被测物是易挥发、不稳定的化学物质,在试验过程中的挥发或分解会造成被测物浓度的下降,使试验溶液的毒性降低。但也有例外,如农药对硫磷的降解产物对氧磷,其毒性高于对硫磷,因而在试验过程中试验溶液的毒性反有所增加。此外,被测物浓度的下降还同微生物的作用,试验容器的性质,沉淀物和悬浮物的吸附,以及受试生物的吸附等有关。被测物是生化需氧量(BOD)高的废水或耗氧量大的化学物质,能消耗试验溶液中的溶解氧,有时甚至会使溶解氧减少到零。受试生物的代谢产物(二氧化碳、氨等)如不及时排除,也会使水质恶化。溶解氧的减少和代谢产物的积累,不仅影响毒物的毒性,而且危害受试生物。因此在进行生物测试时,不仅要设法保持被测物浓度和试验溶液水质的稳定,还要严格控制其他试验条件。    测试方法  在静水式生物测试中,试验溶液如果通过稀释水的预先曝气,仍不能满足试验对溶解氧的要求,可采用有控制的人工充气来补充溶解氧,或定期更新试验溶液。定期更新试验溶液的生物测试称为换水式生物测试,或称为换水式试验,即每隔一定时间将容器中的试验溶液吸出,加入新配制的试验溶液,或将受试生物转入新配制的浓度相同的试验溶液中。更换试验溶液的时间视被测物的性质而定,一般24小时更换一次。如果在24小时内被测物浓度仍有明显下降,或者溶解氧低于饱和度50%时,更新溶液的时间可适当缩短。换水式生物测试除能提供受试生物所需的溶解氧化,还能保持被测物浓度大致稳定和防止水质恶化,因而是静水式生物测试常用的测试方法。    优缺点  19世纪末就有人用静水式生物测试方法测定化学物质和工业废物对鱼类的毒性,但直到20世纪50年代才制定测试的详细程序。与流水式生物测试相比较,静水式生物测试方法简单易行,费用低廉,用水量和废水量均较少,因而至今仍被广泛采用。但是,静水式生物测试只适用于测定不易挥发、相对稳定而又耗氧量不大的污染物,以及BOD不高的工业废水的急性毒性。    参考书目   M.C.Rand et al.,Standard Methods for the Examination of Water and Wastewater,14th ed., Bru-E1.GraphicInc.,Springfield,1979.
